Epigenetics of cardiomyopathies: the next frontier.

Heart Fail Rev

January 2025

Thoraxcenter, Department of Cardiology, Cardiovascular Institute. Erasmus MC - University Medical Center Rotterdam, Office RG-431Dr. Molewaterplein 40, 3015 GD, Rotterdam, The Netherlands.

Cardiomyopathies (CMP) are a diverse group of myocardial diseases that cause structural, functional, and pathological changes to the heart. Alterations at the molecular level associated with the clinical phenotype and progression of CMPs cannot be solely explained by the genetic mutations, even in inherited cardiomyopathies. Epigenetics and environmental factors are likely to significantly modify the clinical manifestations of CMPs, resulting in variable clinical expression and different age-related penetrance.

The Effects of Brodalumab on the Fungal Microbiome in Patients with Psoriasis.

Int J Mol Sci

September 2024

Department of Dermatology and Venereology, Institute of Clinical Sciences, Sahlgrenska Academy, University of Gothenburg, 413 45 Gothenburg, Sweden.

Article Synopsis
  • The gut microbiota is crucial for immune function, and its imbalance (dysbiosis) is associated with systemic inflammation, affecting conditions like psoriasis and depression.
  • This study investigates how switching psoriasis patients from TNF-α inhibitors to brodalumab affects their gut fungal microbiota, using fecal samples and advanced sequencing techniques.
  • Results indicate that while the dominant fungal group was Ascomycota, switching treatments did not significantly change fungal composition, despite clinical improvements, suggesting brodalumab may not impact fungal communities in psoriasis patients.

Article Synopsis
  • - The study examines nitrate levels and microbial communities in deep karst carbonate rocks, using borehole samples collected up to 350 meters deep from proposed railway tunnel sites and analyzed through 16S rRNA sequencing.
  • - Results showed low microbial diversity, indicating the karst environment is relatively pristine, with Pseudomonadota and Gammaproteobacteria being the most prevalent groups, while local diversity hotspots exist independently of depth.
  • - Low nitrate levels in deep karst suggest minimal pollution, linked to natural microbial processes rather than external influences, although increased nitrate is likely due to human activity, and factors beyond depth also affect microbial community structure.

The combination of metformin and the peroxisome proliferator-activated receptors (PPAR) agonists offers a promising avenue for managing type 2 diabetes (T2D) through their potential complementary mechanisms of action. The results from randomized controlled trials (RCT) assessing the efficacy of PPAR agonists plus metformin versus metformin alone in T2D are inconsistent, which prompted the conduct of the systematic review and meta-analysis. We searched MEDLINE and EMBASE from inception (1966) to March 2023 to identify all RCTs comparing any PPAR agonists plus metformin versus metformin alone in T2D.
