Variants in MICOS10 Identified by Whole Genome Sequencing and RNA Sequencing in a New Type of Hepatocerebral Mitochondrial DNA Depletion Syndrome.

Liver Int

January 2025

Diagnostics and Therapeutic of Intractable Diseases, Intractable Disease Research Center, Graduate School of Medicine, Juntendo University, Tokyo, Japan.

Article Synopsis
  • * In a case study, whole genome sequencing revealed a variant in the MICOS10 gene that was associated with mitochondrial hepatopathy and mitochondrial DNA depletion, highlighting specific genetic alterations in the patient.
  • * The findings suggest that loss of MICOS10 can lead to serious conditions like hepatopathy and neuropathy, drawing parallels with diseases linked to another gene in the MICOS complex, MICOS13.

Article Synopsis
  • Gallibacterium anatis and Enterococcus cecorum are bacteria linked to opportunistic infections in poultry and occasionally cause infections in immunocompromised individuals from contaminated food.
  • A 78-year-old Japanese man with acute cholangitis was found to have a coinfection of these bacteria, which were identified as highly virulent and multidrug-resistant strains through genome sequencing.
  • The case highlights that a common bile duct stone can pose a risk for infections by these pathogens even in healthy individuals, underscoring the need for monitoring these bacteria in animals and understanding their effects on human health.

Epigenetic dynamics of partially methylated domains in human placenta and trophoblast stem cells.

BMC Genomics

November 2024

Division of Bioinformatics, Medical Institute of Bioregulation, Kyushu University, Fukuoka, 812- 8582, Japan.

Background: The placenta is essential for nutrient exchange and hormone production between the mother and the developing fetus and serves as an invaluable model for epigenetic research. Most epigenetic studies of the human placenta have used whole placentas from term pregnancies and have identified the presence of partially methylated domains (PMDs). However, the origin of these domains, which are typically absent in most somatic cells, remains unclear in the placental context.

Division of Rheumatology, Department of Internal Medicine, Keio University School of Medicine, Tokyo, and Saitama Medical University, Saitama, Japan.

Objectives: To describe safety and efficacy of filgotinib 200 or 100 mg (FIL200/FIL100) in Japanese patients with rheumatoid arthritis in a long-term extension (LTE; NCT03025308).

Methods: Patients who completed any of three parent studies (NCT02889796: inadequate response [IR] to methotrexate [MTX]; NCT02873936: IR to biologic disease-modifying antirheumatic drugs; NCT02886728: MTX-naïve) without rescue therapy could enter the LTE; patients taking FIL continued their dosage, and those who received comparators were rerandomised to FIL200 or FIL100. This analysis includes week 156 interim results.

The promising diagnostic performance of molecular testing for syphilis using saliva and urine samples has been reported; however, further evaluation of its possible application for diagnosis and molecular surveillance is required. In addition, the development of a rapid and easy-to-perform molecular test for syphilis is important for its use in the clinical setting. We comprehensively evaluated the diagnostic and surveillance performance of two novel loop-mediated isothermal amplification (LAMP) assays using saliva and urine samples.

Aim: Lynch syndrome (LS) is a dominantly inherited syndrome characterized by an increased risk for LS associated tumors such as colorectal cancer (CRC) and gastric cancer (GC). However, the clinical benefit of surveillance for GC remains unclear while it has already been recommended for CRC. This study aimed to elucidate the clinical features of GC in Japanese individuals with LS, and the risk of developing multiple GCs to build regional-tailored surveillance programs in LS patients with GC.

  • Non-Hodgkin lymphoma (NHL) can resemble soft tissue tumors, making pre-biopsy diagnosis difficult; this study aimed to determine the effectiveness of serum soluble interleukin-2 receptor (sIL-2R) levels and MRI in diagnosing soft tissue NHL.* -
  • Researchers analyzed 36 cases of confirmed soft tissue NHL alongside 48 control cases and found that combining MRI results (homogeneous appearance) and elevated sIL-2R levels effectively identified soft tissue NHL with high sensitivity and specificity.* -
  • The study concluded that using both MRI and sIL-2R measurements together provides a reliable method for predicting soft tissue NHL, enhancing diagnostic accuracy compared to conventional tumor evaluations.*

Article Synopsis
  • This study investigated the use of amylase levels in drainage fluid (DFA) to detect anastomotic leakage (AL) early and non-invasively in patients undergoing low anterior resection (LAR) for rectal cancer.
  • The study analyzed drainage fluid samples from 120 patients post-surgery, finding that DFA levels were significantly higher in those with AL, particularly on postoperative days 3 and 5.
  • The results indicated that DFA levels on day 5 offered excellent diagnostic accuracy for detecting AL, with high sensitivity and specificity, suggesting that this method could be a simple and cost-effective way to detect complications early in these patients.

Article Synopsis
  • The study investigates the effectiveness of catheter ablation in eliminating idiopathic ventricular arrhythmias from different sites in the heart.
  • It analyzes data from 58 patients and finds that overall, 72% had successful ablation, with different success rates linked to two specific sites: ECS and endo-EAS.
  • Successful ECS ablation is associated with shorter distances to the activation site, while successful endo-EAS ablation is linked to earlier electrical signals and shorter intervals between activation sites.

Medication-related osteonecrosis of the jaw (MRONJ) caused by bisphosphonates (BPs) and denosumab (Dmab) is still a major oral adverse event caused by cancer treatment and may be under-recognized by private dental services. With the aim of reducing the prevalence of MRONJ, this study, which is a pilot study for a planned future large-scale survey, compared knowledge about MRONJ between dental hygienists (DHs) in private dental clinics and those in cancer center hospitals. We conducted a questionnaire-based survey regarding MRONJ between 1 November 2023 and 31 January 2024 on DHs at a cancer base hospital and a private clinic in Saitama, Japan.

Myofibroblastomas are benign mesenchymal tumors that frequently occur in the groin. They show variable morphology, and the differential histopathological diagnoses are broad, including lipomatous to myxoid tumors. In addition, both pathological and imaging findings may be complex, which makes diagnosis challenging.

Objective: This study aimed to evaluate the diagnostic performance of 99m Tc-hydroxymethylene diphosphonate ( 99m Tc-HMDP) imaging for cardiac amyloidosis and to demonstrate changes in cardiac uptake of 99m Tc-HMDP after tafamidis treatment.

Methods: Seventy-five patients with suspected cardiac amyloidosis who underwent 99m Tc-HMDP imaging were included. We compared visual Perugini grades and semiquantitative heart-to-contralateral (H/CL) area ratios, myocardial maximum standardized uptake value (SUVmax), and peak of SUV (SUVpeak) between cardiac transthyretin amyloidosis (ATTR) and amyloid light-chain amyloidosis (AL).
