Biliary intraepithelial neoplasm (BilIN) and intraductal papillary mucinous neoplasm of the bile duct (IPMN-B) are recognized as the two main precursors to biliary carcinoma. Surgical resection is the mainstay of treatment, with pancreaticoduodenectomy for extrahepatic biliary duct disease involving the pancreatic or distal portion of the bile duct, and bile duct and liver resection for perihilar and intrahepatic bile duct involvement. For diffuse IPMN-B with involvement of the entirety of the biliary epithelium of both intrahepatic and extrahepatic biliary systems, there is no well-documented consensus on treatment due to its rarity.

BMC Nephrol

October 2024

Department of Pediatrics, Division of Nephrology, Saint Louis University, 1465 S Grand Blvd, St. Louis, MO, 62104, USA.

Background: Tuberous Sclerosis complex (TSC) is a multisystemic neurocutaneous genetic condition with high rates of morbidity and mortality from subependymal giant cell astrocytoma (SEGA), renal angiomyolipoma, and renal cyst complications. Everolimus is an inhibitor for mTORC1 and is currently used to treat TSC for its main role in rapidly reducing SEGA volume and seizure burden, although mainly studied in the adult population. It has also been shown to stabilize estimated glomerular filtration rate and reduce renal angiomyolipoma size in the adult population.

Chimeras Derived from a P2Y Receptor Antagonist and UDP-Sugar Agonists for Potential Treatment of Inflammation.

ACS Pharmacol Transl Sci

October 2024

Molecular Recognition Section, Laboratory of Bioorganic Chemistry, National Institute of Diabetes and Digestive and Kidney Diseases, National Institutes of Health, Bethesda, Maryland 20892, United States.

Tethered glycoconjugates of a naphthalene- and piperidine-containing antagonist of the P2Y receptor (PPTN) were synthesized, and their nM receptor binding affinity was determined using a fluorescent tracer in hP2YR-expressing whole CHO cells. The rationale for preparing mono- and disaccharide conjugates of the antagonists was to explore the receptor binding site, which we know recognizes a glucose moiety on the native agonist (UDP-glucose), as well as enhance aqueous solubility and pharmacokinetics, including kidney excretion to potentially counteract sterile inflammation. Glycoconjugates with varied linker length, including PEG chains, were compared in hP2YR binding, suggesting that an optimal affinity (IC, nM) in the piperidine series was achieved for triazolyl -linked glucose conjugates having one (, MRS4872, 3.

Article Synopsis
  • The study aimed to determine how different durations of intraoperative radiofrequency ablation affect the health and characteristics of bursal cells collected during rotator cuff surgery in patients.
  • Six healthy patients participated, and bursal tissue was subjected to varying levels of ablation to assess cell viability, growth potential, and phenotypes.
  • Findings showed that longer ablation times led to reduced viability of the cells, but the overall characteristics of the cells remained consistent across different treatment groups, indicating that while ablation affects cell survival, it does not significantly alter cell properties.
