Introduction: White matter hyperintensities (WMHs) are an important imaging marker for cerebral small vessel diseases, but their risk factors and cognitive associations have not been well documented in populations of different ethnicities and/or from different geographical regions.

Methods: We investigated how WMHs were associated with vascular risk factors and cognition in both Whites and Asians, using data from five population-based cohorts of non-demented older individuals from Australia, Singapore, South Korea, and Sweden ( = 1946). WMH volumes (whole brain, periventricular, and deep) were quantified with UBO Detector and harmonized using the ComBat model.

SSPC, the, Science Foundation Ireland Research Centre for Pharmaceuticals, School of Chemical Sciences, Dublin City University, Glasnevin, Dublin 9, Dublin, Ireland.

Artificial metallo-nucleases (AMNs) are promising DNA damaging drug candidates. Here, we demonstrate how the 1,2,3-triazole linker produced by the Cu-catalysed azide-alkyne cycloaddition (CuAAC) reaction can be directed to build Cu-binding AMN scaffolds. We selected biologically inert reaction partners tris(azidomethyl)mesitylene and ethynyl-thiophene to develop TC-Thio, a bioactive C -symmetric ligand in which three thiophene-triazole moieties are positioned around a central mesitylene core.

Introduction: Plasma neurofilament light chain (NfL) is a potential biomarker for neurodegeneration in Alzheimer's disease (AD), ischemic stroke, and non-dementia cohorts with cerebral small vessel disease (CSVD). However, studies of AD in populations with high prevalence of concomitant CSVD to evaluate associations of brain atrophy, CSVD, and amyloid beta (Aβ) burden on plasma NfL are lacking.

Methods: Associations were tested between plasma NfL and brain Aβ, medial temporal lobe atrophy (MTA) as well as neuroimaging features of CSVD, including white matter hyperintensities (WMH), lacunes, and cerebral microbleeds.

Article Synopsis
  • The study investigates the impact of peripheral artery disease (PAD) on the outcomes of transcatheter mitral valve repair (TMVr) in heart failure (HF) patients with severe mitral regurgitation, comparing results with guideline-directed medical therapy (GDMT) alone.
  • Among 614 patients, 109 had PAD; findings showed that PAD was linked to higher mortality but did not affect hospitalization rates for heart failure.
  • TMVr significantly reduced mortality for patients without PAD, but not for those with PAD; however, it consistently decreased hospitalization rates and improved health status and exercise capacity for all patients regardless of PAD status.

Background: Although most nondisplaced or minimally displaced femoral neck fractures are routinely treated with internal fixation, high rates of secondary surgical procedures are common, especially in the elderly population. Primary arthroplasty in elderly patients has been proposed as an alternative treatment to reduce the need for a secondary surgical procedure. The objective of this study was to describe the rate of conversion to arthroplasty within 5 years after internal fixation of nondisplaced femoral neck fractures in patients ≥60 years of age.

Article Synopsis
  • A study analyzed the effectiveness of the modified ACC/AHA lesion classification as a way to predict short- and long-term outcomes in patients undergoing percutaneous coronary intervention (PCI) with second-generation drug-eluting stents.
  • Data from 8,516 patients revealed that those with complex lesions (B2/C) experienced higher instances of target lesion failure (TLF) compared to those with noncomplex lesions (A/B1) at multiple follow-up points (30 days, 1 year, and 5 years).
  • The findings suggest that the historical ACC/AHA classification system remains relevant for predicting clinical outcomes after PCI, particularly highlighting worse outcomes for complex lesions after 5 years.
