The description of a cell-free soluble anti-tumour factor by Carswell et al. in 1975 (Proc Natl Acad Sci USA, 72: 3666-3670) was followed by a long series of experimental and clinical investigations into the role of cell-free mediators in cancer immunotherapy. These investigations included research on the effects of macrophage-derived eicosanoids (cycloxygenase and lipoxygenase derivates of arachidonic acid) and of monokines such as tumour necrosis factor-alpha, interleukin-1 and granulocyte-monocyte-macrophage-colony stimulating factor) and of lymphocyte products: interleukins and interferons.

Human monocytes, co-incubated for 7 days in culture with GM-CSF or IL-3 but not with IFN-gamma, exerted a variable schistosotnulicidal effect on Schistosoma mansoni parasites when grown in 96-well round-bottomed plates but not in flat-bottomed plates. Addition of LPS or IFN-gamma or both, for the last 48 h did not enhance the cidal effect. Addition of LPS but not IFN-gamma to the pre-incubated cells with GM-CSF or IL-3 markedly stimulated TNF-alpha production by the cells but not their cidal activity.
