Cellular protein homeostasis in the lungs is constantly disrupted by recurrent exposure to various external and internal stressors, which may cause considerable protein secretion pressure on the endoplasmic reticulum (ER), resulting in the survival and differentiation of these cell types to meet the increased functional demands. Cells are able to induce a highly conserved adaptive mechanism, known as the unfolded protein response (UPR), to manage such stresses. UPR dysregulation and ER stress are involved in numerous human illnesses, such as metabolic syndrome, fibrotic diseases, and neurodegeneration, and cancer.

Ischemia/reperfusion (I/R) injury is a serious pathologic event that occurs due to restriction in blood supply to an organ, followed by hypoxia. This condition leads to enhanced levels of pro-inflammatory cytokines such as IL-6 and TNF-α, and stimulation of oxidative stress via enhancing reactive oxygen species (ROS) levels. Upon reperfusion, blood supply incz reases, but it deteriorates condition and leads to the generation of ROS, cell membrane disruption and finally, cell death.

Breast cancer is one of the main reasons of women's mortality. This study aimed to improve delivery of lomustine as a chemotherapeutic agent and to increase its uptake by U87-MG cancer cells via synthesizes LN-FA-PG-SPIONs (lomustine loaded polyglycerol coated superparamagnetic iron oxide nanoparticles conjugated with folic acid). Nanoparticles were synthesized by thermal decomposition method and characterized using TEM (transmission microscope), FTIR (Fourier transform infrared spectroscopy), and VSM (vibrating sample magnetometer). Lomustine release from nanoparticles was determined by dialysis-bag diffusion technique.

Doxorubicin (DOX) is a well-known chemotherapeutic agent extensively applied in the field of cancer therapy. However, similar to other chemotherapeutic agents such as cisplatin, paclitaxel, docetaxel, etoposide and oxaliplatin, cancer cells are able to obtain chemoresistance that limits DOX efficacy. In respect to dose-dependent side effect of DOX, enhancing its dosage is not recommended for effective cancer chemotherapy.

In this work, for the first time, a novel pH-sensitive biocompatible multifunctional nanocarrier was fabricated by the combination of MgAl-layered double hydroxide, MnO nanoparticles, N-graphene quantum dot and polyaniline (PANI/N-GQD/MO/LDH) for doxorubicin (DOX) delivery in breast cancer cells. Electrochemical techniques, including cyclic voltammetry (CV) and differential pulse voltammetry (DPV), were employed for proving the surface modification process. The integration of polyaniline on the surface of the nanocarrier provides ultrahigh DOX encapsulation up to 90% and possesses a slow-release behavior (4% after 72 h) under normal physiological conditions.

Ubiquitination is a multi-step enzymatic process that involves the marking of a substrate protein by bonding a ubiquitin and protein for proteolytic degradation mainly via the ubiquitin-proteasome system (UPS). The process is regulated by three main types of enzymes, namely ubiquitin-activating enzymes (E1), ubiquitin-conjugating enzymes (E2), and ubiquitin ligases (E3). Under physiological conditions, ubiquitination is highly reversible reaction, and deubiquitinases or deubiquitinating enzymes (DUBs) can reverse the effect of E3 ligases by the removal of ubiquitin from substrate proteins, thus maintaining the protein quality control and homeostasis in the cell.
