ATP synthase of E. coli: F-ATPase activity is functionally decoupled from the proton-transporting complex (F) by tributyltin.

Department of Biochemistry & Molecular Biology, SUNY Upstate Medical University, 750 E Adams St, Syracuse, NY, 13210, USA. Electronic address:

The F-type ATP synthase/ATPase (FF) is important for cellular bioenergetics in eukaryotes and bacteria. We recently showed that venturicidins, a class of macrolides that inhibit the proton transporting complex (F), can also induce time-dependent functional decoupling of F-ATPase from F on membranes from Escherichia coli and Pseudomonas aeruginosa. This dysregulated ATPase activity could deplete bacterial ATP levels and contribute to venturicidin's capacity to enhance the bactericidal action of aminoglycosides antibiotics.

Background: Obstructive sleep apnea (OSA) is more common in patients with multiple sclerosis (MS) than in the general population, which suggests MS may predispose patients to OSA. However, the relationships between MS treatment, disease activity, disease severity, fatigue, and OSA are unknown.

Objectives: To evaluate the connections between OSA risk, MS fatigue, and MS severity, controlling for well-established risk factors for OSA in the general population.

Spinal alignment and surgical correction in the aging spine and osteoporotic patient.

N Am Spine Soc J

September 2024

Department of Orthopedic Surgery and Department of Pediatrics, SUNY Upstate Medical University, 705 E. Adams St., Syracuse, NY 13201, United States.

Article Synopsis
  • The aging spine can present complex surgical challenges due to issues like spinal alignment and quality of life, especially in elderly patients who may have osteoporosis and frailty.
  • A literature review highlighted factors affecting spinal alignment, such as flexibility and bone health, and emphasized the importance of thorough preoperative assessments and radiographic evaluations to guide surgical planning.
  • Personalized surgical strategies should be developed based on individual patient conditions and health, with the potential use of anabolic agents to improve bone quality and reduce complications related to surgery.

Plasma cell-rich acute rejection (PCAR), a relatively rare subtype of acute allograft rejection, is usually associated with a significantly lower treatment response rate and a higher graft failure rate. PCAR is characterized by the presence of more than 10% of plasma cells out of all graft infiltrating cells, with approximately 40%-60% of PCAR resulting in graft failure within a year. Currently, there is no gold standard for the effective treatment of PCAR.

Background: Squamous cell carcinoma (SCC) of presumed lung origin (PLO) is now the second most frequent histologic subtype of non-small cell carcinoma after adenocarcinoma. The use of clinic-genomic correlation provided by comprehensive genomic profiling (CGP) can revise clinicopathologic diagnoses of presumed primary lung SCC (PLO-SCC) to diagnoses of metastatic SCC of cutaneous origin (C-SCC).

Design: A total of 10 146 samples of clinically advanced PLO-SCC (84% known Stage IV) passed QC metrics and were designated as PLO-SCCs by review of test requisition forms, clinical notes, and pathology reports.

Background: The effectiveness of the clinical outcome of CN (Cytoreductive Nephrectomy) in cases of mccRCC (Metastatic Clear Cell Renal cell Carcinoma) is still uncertain despite two trials, SURTIME and CARMENA. These trials, conducted with Sunitinib as the standard treatment, did not provide evidence supporting the use of CN.

Methods: We queried the NCDB for stage IV mccRCC patients between the years of 2004 to 2020, who received (immunotherapy) IO with or without nephrectomy.

Aging is a multifaceted process that may lead to an increased risk of developing cancer. Artificial intelligence (AI) applications in clinical cancer research may optimize cancer treatments, improve patient care, and minimize risks, prompting AI to receive high levels of attention in clinical medicine. This systematic review aims to synthesize current articles about the effectiveness of artificial intelligence in cancer treatments for older adults.

Schizophrenia (SCH) is a mental disorder that requires long-term antipsychotic treatment. SCH patients are thought to have an increased sensitivity to stress. The dysregulation of the hypothalamic-pituitary-adrenal (HPA) axis, observed in SCH, could include altered levels of glucocorticoids, glucocorticoid receptors (GRs), and associated proteins.

Multiple myeloma is a heterogeneous condition characterized by the proliferation of monoclonal B-cells, for which there is currently no curative treatment available. Relapses are, unfortunately, common after first-line treatment. While the prognosis for relapsed refractory multiple myeloma is generally poor, advances in the treatment of relapsed or refractory multiple myeloma offer hope.

Serotonin has been implicated in the neurobiology of attention-deficit/hyperactivity disorder (ADHD) due to its association with impulsivity, attention, and emotional regulation. Many compounds with serotonergic properties have been evaluated in ADHD, but few have been approved by regulatory authorities. Utilizing a search of public databases, we identified interventions studied in ADHD.

Introduction: Acute lymphoblastic leukaemia (ALL) is the most common type of childhood leukaemia with effective chemotherapeutic treatment. However, obesity has been associated with higher ALL chemoresistance rates and lower event-free survival rates. The molecular mechanism of how obesity promotes chemotherapy resistance is not well delineated.

  • The USMLE is a crucial exam for allopathic physicians in the US, with its content developed by the NBME, but the specialties of the taskforce members contributing to the exam have not been well-documented.
  • An analysis of the 2021 USMLE and NBME taskforce revealed significant differences in the primary specialties of test writers compared to the overall population of active physicians in the US, with only nineteen specialties represented among USMLE writers.
  • Internal Medicine was the most represented specialty among USMLE test writers, highlighting a potential imbalance that could affect the fairness and comprehensiveness of the exam content, suggesting the need for further research.

Internal validation of modified Mirels' scoring system for pathologic femur fractures.

BMC Musculoskelet Disord

September 2024

Department of Orthopedic Surgery, SUNY Upstate Medical University, 750 East Adams Street, Syracuse, NY, 13210, USA.

Background: The proximal femur is a common site of bone metastasis. The Mirels' score is a frequently utilized system to identify patients at risk for pathologic fracture and while it has consistently demonstrated strong sensitivity, specificity has been relatively poor. Our group previously developed a Modified Mirels' scoring system which demonstrated improved ability to predict cases at risk of fracture in this patient population through modification of the Mirels' location score.

  • The study aimed to assess how the neutrophil to lymphocyte ratio (NLR) impacts the outcomes of patients with penile squamous cell carcinoma (PSCC).
  • A meta-analysis was performed, analyzing 14 studies to understand the relationship between NLR levels and patient survival, utilizing established quality assessment criteria.
  • Results indicated that higher NLR levels were linked to worse prognosis and lower survival rates in PSCC patients, confirming NLR's important role in diagnosis and prognosis despite no link to tumor size or stage.

  • Nirsevimab is a long-lasting monoclonal antibody designed to prevent RSV-related respiratory issues in vulnerable infants and children, and its effectiveness was tested against the standard treatment, palivizumab, in a clinical trial called MEDLEY.
  • The trial included two RSV seasons where participants received either nirsevimab or palivizumab, with ongoing assessments of RSV infections and antibody responses through nasal swabs.
  • The results showed that while certain substitutions in RSV isolates developed resistance to palivizumab, no changes were found that affected nirsevimab's ability to neutralize RSV, indicating its potential superiority in preventing RSV infections.
