Upregulation of Urinary Alpha-2-HS Glycoprotein During Pregnancy of Murrah Buffaloes.

Urine samples were systematically collected from inseminated Murrah buffaloes (Bubalus bubalis) on days 0, 7, 14, 21 and 28 (with day 0 representing the day of artificial insemination). Following confirmation of pregnancy via trans rectal palpation 45 days of insemination, the animals were categorised into pregnant and non-pregnant groups (n = 10 each). The urine samples on 0, 7, 14, 21 and 28 days of pregnant and one sample from non-pregnant preferably collected on 28th day was used for SDS-PAGE after diafiltration.

The present study was conducted to elucidate (1) the influence of kisspeptin (KP) on the development of preantral follicles (PFs) and (2) evolution of KP receptor gene () expression during ovarian follicular development in sheep. Kisspeptin was supplemented (0-100 µg/ml) in the culture medium of PFs for 6 days. The cumulus-oocyte complexes (COCs) from cultured PFs were subsequently matured to metaphase II (MII) for an additional 24 h.

Quantifiable decline in the maternal body temperature during the pre-calving offers the possibilities for predicting the calving that can improve the calving management. As infrared thermography (IRT) is a simple non-contact tool for precise measurement of surface temperature, we investigated the use of IRT to establish thermal signatures around calving in the Murrah buffalo. The IRT of eye, right lateral, left lateral and rear side of udder skin surface temperature (USST) were recorded at 6 h interval from 96 h before the expected date of calving, at the time of calving and 24 h post-calving in Murrah buffaloes (n = 28).

The Signal Transducer and Activator of Transcription 5A (STAT5A) gene involved in activating the transcription of milk protein genes was predicted to be influencing milk production traits. The present study was undertaken to investigate the suitability of the polymorphism of STAT5A as a marker for milk traits in Ongole, crossbred cattle and Murrah buffaloes from Southern India. The objective of this study was to test the hypothesis that Leptin induced in vitro growth in preantral follicles in sheep involves modulation of P450 aromatase expression and steroidogenesis. Accordingly, the expression of P450 aromatase gene was studied in the cumulus cells and oocytes isolated from different stages of preantral follicles (PFs') grown in vivo, cultured in TCM 199B, TCM 199B + Leptin (10 ng/ml) (TCM199BL) or a standard PF culture medium supplemented with Leptin (10 ng/ml) (SML). Ovarian follicles grown in vivo or in SML expressed P450 aromatase both in cumulus cells and oocytes at all the development stages.

Expression of Kit ligand (KL) and insulin-like growth factor binding protein (IGFBP3) genes was studied at different in vivo and corresponding in vitro stages of development of the ovarian follicles in sheep. The expression of both KL and IGFBP3 was significantly higher in the primordial follicles relative to any other stage of development. Compared to the other stages, the KL expression in the cumulus cells from in vivo grown large antral follicles and that of IGFBP3 in COCs' isolated from large antral follicles matured in vitro for 24 hr were significantly higher.

The compounds tested were neem coated silver nanoparticles (N-Ag NPs), deltamethrin neem coated silver nanoparticles (DN-Ag NPs), 2, 3 dehydrosalannol (2,3 DHS), 2, 3 DHS coated silver nanoparticles (2, 3-DHS-Ag NPs), Quercetin dihydrate (QDH) and QDH coated silver nanoparticles (QDH-Ag NPs).
