Thyroid transcription factor-1 expression in lung neuroendocrine tumours: a gender-related biomarker?

Endocrine

February 2024

Endocrinology Unit, Department of Clinical and Molecular Medicine, Sant'Andrea Hospital, Sapienza University of Rome, ENETS Center of Excellence, Rome, Italy.

Purpose: Thyroid transcription factor-1 (TTF-1) assessed by immunohistochemistry (IHC) is a specific biomarker for lung adenocarcinoma, and is commonly used to confirm the pulmonary origin of neuroendocrine tumours (NET). The majority of the available data suggest that TTF-1 is favourable prognostic biomarker for lung adenocarcinomas, whereas its role is more conflicting for lung NET. The main aim of this multicenter retrospective study was to investigate the potentially relevant associations between TTF-1 biomarker and clinical and pathological features of the study population, as well as determine TTF-1 prognostic effect on the clinical outcome of the patients.

Article Synopsis
  • Modern non-Africans have 1-3% Neandertal DNA due to interbreeding events that occurred 50,000-60,000 years ago, which may affect traits linked to diseases like pancreatic ductal adenocarcinoma (PDAC).
  • In this study, researchers examined how specific Neandertal-related genetic variations (aSNPs) correlate with PDAC risk in European and East Asian populations, using data from over 200,000 individuals.
  • While no significant link was found in Europeans, a specific allele in East Asians was associated with a 35% increased risk of developing PDAC, suggesting only a limited role of Neandertal genes in this cancer's risk.

Article Synopsis
  • Heart failure is a serious side effect of doxorubicin (DOX) in cancer patients, and this study investigates the role of the MST1 kinase in DOX-induced heart damage.
  • Researchers used mice with normal MST1 and those with a modified version that can't activate (dominant-negative) to analyze the effects of DOX treatment, finding that MST1 activation contributes to heart injury.
  • The study concludes that inhibiting MST1 can protect against DOX-induced heart damage by preventing the downregulation of SIRT3, a protective protein, which was also found altered in heart tissue from cancer patients receiving DOX.

Exhaled nitric oxide (NO) production, upregulated by inflammatory cytokines and mediators in central and peripheral airways, can be easily and non-invasively detected in exhaled air in asthma and other respiratory conditions as a promising tool for disease monitoring. The American Thoracic Society and European Respiratory Society released recommendations that standardize the measurement of the fractional exhaled NO (FeNO). In asthma, increased FeNO reflects eosinophilic-mediated inflammatory pathways and, as a biomarker of T2 inflammation can be used to identify asthma T2 phenotype.
