Microbiota and luminal components may affect epithelial integrity and thus participate in the pathophysiology of colon cancer (CC) and inflammatory bowel disease (IBD). Therefore, we aimed to determine the effects of fecal luminal factors derived from patients with CC and ulcerative colitis (UC) on the colonic epithelium using a standardized colon-derived two-dimensional epithelial monolayer. The complex primary human stem cell-derived intestinal epithelium model, termed RepliGut Planar, was expanded and passaged in a two-dimensional culture which underwent stimulation for 48 h with fecal supernatants (FS) from CC patients ( = 6), UC patients with active disease ( = 6), and healthy subjects (HS) ( = 6).

Bacterial contamination and greenhouse gas emissions: A randomised study of reuse versus single-use of infusion-set components for intravenous anaesthesia.

Eur J Anaesthesiol

December 2024

From the Department of Anaesthesia, SV Hospital Group, Institute of Clinical Sciences at the Sahlgrenska Academy, University of Gothenburg, Gothenburg (SLK, MVG, PWM), Department of Infection Prevention and Control, Södra Älvsborg Hospital, Borås (JEW), Department of Infectious Disease, Institute of Biomedicine at the Sahlgrenska Academy, University of Gothenburg, Gothenburg (JEW), and Anaesthesiology and Intensive Care, Department of Clinical Sciences Lund, Lund University, Lund, Sweden (PB).

Background: Reusing anaesthesia infusion-set components may reduce the climate impact from plastic waste and discarded medications. Infusion-set contents can be shielded from patient contact by single use of an infusion line fitted with dual antireflux valves, preventing retrograde entry of microorganisms, and eliminating the risk for patient-to-patient cross-contamination. However, infusion-set contamination from compromised aseptic handling could affect quality of care.

Repurposing of FDA-approved drugs is a quick and cost-effective alternative to de novo drug development. Here, we identify genes involved in bortezomib sensitivity, predict cancer types that may benefit from treatment with bortezomib, and evaluate the mechanism-of-action of bortezomib in breast cancer (BT-474 and ZR-75-30), melanoma (A-375), and glioblastoma (A-172) cells in vitro. Cancer cell lines derived from cancers of the blood, kidney, nervous system, and skin were found to be significantly more sensitive to bortezomib than other organ systems.

Article Synopsis
  • Multiple myeloma (MM) is a type of cancer affecting plasma cells, with a significant genetic component that is not fully understood.
  • A large genome-wide study identified 35 risk loci related to MM, including 12 new ones, and revealed two main inherited risk factors: longer telomeres and higher levels of B-cell maturation antigen (BCMA) and interleukin-5 receptor alpha (IL5RA) in the blood.
  • The genetic variant rs34562254-A increases the risk of MM by enhancing B-cell responses, contrasting with loss-of-function variants in TNFRSF13B that lead to B-cell immunodeficiency.
