Biomolecules

December 2024

Division of Experimental and Molecular Psychiatry, Department of Psychiatry, Psychotherapy and Preventive Medicine, LWL University Hospital, Ruhr University Bochum, 44801 Bochum, Germany.

Exposure to stress during early and late childhood can lead to long-lasting neurobiological and behavioral impairments. Although sensitive periods for stress exposure are well established, less is known about the trajectory of induced alterations throughout development. In this study, we investigated the impact of maternal separation (MS), social isolation, and their combination on anxiety-like behavior and gene expression across developmental stages.

Background: Machine learning (ML) is increasingly used to predict clinical deterioration in intensive care unit (ICU) patients through scoring systems. Although promising, such algorithms often overfit their training cohort and perform worse at new hospitals. Thus, external validation is a critical - but frequently overlooked - step to establish the reliability of predicted risk scores to translate them into clinical practice.

Auranofin induces disulfide bond-mimicking S-Au adducts in protein thiol pairs.

J Biol Chem

January 2025

Microbial Biochemistry, Faculty of Medicine, Ruhr University Bochum, 44780 Bochum, Germany. Electronic address:

Auranofin is an inhibitor of human thioredoxin reductase, clinically used in the treatment of rheumatoid arthritis. More recently, it has been shown to possess strong antibacterial activity. Despite the structural dissimilarity and the independent evolutionary origins of human thioredoxin reductase and its bacterial counterpart (TrxB), inhibition of bacterial thioredoxin reductase is often suggested to be a major factor in auranofin's antibacterial mode of action.

Article Synopsis
  • New quinizarin-Au(I)-NHC complexes were developed and fully characterized, demonstrating effective growth inhibition in HeLa cervical cancer cells with IC values between 2.4 and 5.3 μM.
  • Cytotoxicity studies showed that complex 2 b could overcome anthracycline resistance in K562 leukemia cells and worked synergistically with doxorubicin against both sensitive and resistant leukemia cells.
  • The study highlighted that localizing these complexes to mitochondria was crucial for their antiproliferative effects and ability to counteract drug resistance, rather than just overall cytotoxicity.

Intracellular recycling via autophagy is governed by post-translational modifications of the autophagy-related (ATG) proteins. One notable example is ATG4-dependent delipidation of ATG8, a process that plays critical but distinct roles in autophagosome formation in yeast and mammals. Here, we aim to elucidate the specific contribution of this process to autophagosome formation in species representative of evolutionarily distant green plant lineages: unicellular green alga Chlamydomonas reinhardtii, with a relatively simple set of ATG genes, and a vascular plant Arabidopsis thaliana, harboring expanded ATG gene families.

Maternal capabilities to engage in sensitive caregiving are important for infant development and mother-infant-interaction, however, can be negatively affected by cortisol due to a stress response. Previous research suggested that cortisol possibly impairs cognitive functions important for caregiving behavior, which potentially leads to less maternal sensitivity. However, studies investigating the influence of cortisol using endocrine parameters on the mother-infant-interaction during the early postpartum are lacking.
