Objective: In this study, we aimed to assess main factors influencing the Valproic Acid (V.Acid) plasma trough levels (C0) and to determine their degree of influence on V.Acid C0 in children with epilepsy who had Therapeutic Drug Monitoring (TDM).

Objective: LRRK2-G2019S is the most frequent mutation in North African Parkinson's disease (PD) patients. Data on its impact on disease progression and treatment response remain elusive. Therefore, we investigated the clinical features, treatments, and complications of PD in Tunisian patients according to their LRRK2-G2019S profile.

Article Synopsis
  • Tacrolimus shows variability in how it works in different individuals and has a small window between effective and harmful doses, with ongoing debate about the effect of a specific genetic variant (CYP3A5 6986A>G) on this variability.
  • A study on Tunisian kidney transplant recipients examined the effects of this SNP on tacrolimus levels, adverse drug reactions (ADRs), and kidney graft outcomes over five years.
  • Results indicated that patients with the CYP3A5*3 variant had higher tacrolimus concentrations and more ADRs, but this genetic variant did not affect the success of the kidney transplant.

Assessment of the influence of Fc-γ receptor polymorphisms on biologics' pharmacokinetics in Tunisian rheumatoid arthritis patients.

Br J Clin Pharmacol

June 2023

Laboratory of Research in Immunology, Renal Transplantation and Immunopathology (LR03SP01), Charles Nicolle Hospital, Tunis El Manar University, Tunis, Tunisia.

Article Synopsis
  • The study investigates whether variations in Fc-γ receptors caused by specific genetic changes (single nucleotide polymorphisms) affect how well therapeutic monoclonal antibodies are cleared from the body and influence the development of anti-drug antibodies.
  • Conducted on Tunisian rheumatoid arthritis patients treated with various biologic drugs, the study measured serum drug levels and the presence of anti-drug antibodies while genotyping patients for specific Fc-γ receptor variations.
  • Results indicate that while most patients had serum drug levels within the therapeutic range, a minority developed anti-drug antibodies, with a notable association between the high affinity FcgR2A R131H receptor and antibody development, prompting recommendations for further research with larger sample sizes.

Is there a ceiling lamotrigine serum level in epileptic children in remission?

University of Tunis El Manar, Faculty of de Medicine of Tunis, National Centre Chalbi Belkahia of Pharmacovigilance, Department of clinical pharmacology, Research Laboratory of Clinical and Experimental Pharmacology (LR16SP02), 1006 Tunis, Tunisia.

Introduction: Seizure control, in patients with epilepsy, is proportionally associated with health-related quality of life. Antiepileptic therapy leads to seizure remission in most cases. However, some patients are resistant to treatment despite achieving high doses which can be explained by interindividual variability of antiepileptic drugs' metabolism.
