Regression interval mapping and multiple interval mapping are compared with regard to mapping linked quantitative trait loci (QTL) in inbred-line cross experiments. For that purpose, a simulation study was performed using genetic models with two linked QTL. Oocytes were selected on the basis of brillant cresyl blue (BCB) staining, used to indicate glucose-6-phosphate dehydrogenase (G6PDH) activity. To re-valuate the hypothesis that growing oocytes are expected to have a high level of active G6PDH, while mature oocytes have low G6PDH activity, cumulus oocyte complexes (COCs) were recovered from slaughterhouse ovaries by slicing the surface of the ovary.

As an alternative to multiple-interval mapping a two-step moment method was recently proposed to map linked multiple quantitative trait loci (QTLs). The advantage of this moment method was supposed to be its simplicity and computational efficiency, especially in detecting closely linked QTLs within a marker bracket, but also in mapping QTLs in different marker intervals. Using simulations it is shown that the two-step moment method may give poor results compared with multiple-interval mapping, irrespective of whether the QTLs are in the same or in different marker intervals, especially if linked QTLs are in repulsion.

The generation of special crosses between different inbred lines such as recombinant inbred strains (RIS) and intermated recombinant inbred populations (IRIP) is being used to improve the power of QTL detection techniques, in particular fine mapping. These approaches acknowledge the fact that recombination of linked loci increases with every generation, caused by the accumulation of crossovers appearing between the loci at each meiosis. This leads to an expansion of the map distance between the loci.

Cyp19, the key gene of oestrogen biosynthesis, is expressed at very different concentrations and from different promoters in bovine granulosa cells (GCs) and in pregnant corpora lutea (CL), respectively. The present study was aimed to investigate if DNA methylation and thus epigenetic mechanisms might play a potential role in the regulation of Cyp19 expression and promoter-specific activity in GCs of cycling versus CL of pregnant cows. It was demonstrated that GCs express high concentrations of promoter-2-derived Cyp19 transcripts whereas CL samples isolated before and after implantation, and at the end of the first trimester, showed very low Cyp19 transcript concentrations, all of them derived from promoter 1.

Prenatal stress has been seen as a reason for reproductive failures in pig offspring mostly originated or mediated by changed maternal functions. To get more information about the order of genes located in Bos taurus (BTA) chromosome 27 segments, supposed to harbor loci influencing clinical mastitis and somatic cell count, and to identify genes that serve as positional candidates for the mentioned traits, we constructed a high-resolution, comparative, and comprehensive gene map for BTA27. The map includes 57 loci in a 5000-rad cattle-hamster whole genome radiation hybrid panel supported by 50 syntenic assignments in a cattle-murine somatic hybrid cell panel. Thirty-eight new loci (36 genes, 2 microsatellites) together with repeated mappings of 5 genes and 7 microsatellites and integration of existing data from 7 microsatellites were used to generate a comprehensive RH5000 map.

In an intercross between the high-body-weight-selected mouse line NMRI8 and the inbred line DBA/2, we analyzed genetic effects on growth during the suckling period and after weaning during the juvenile phase of development. QTL mapping results indicated that a switch of gene activation might occur at the age of three weeks when animals are weaned. We found QTLs for body weight with major effects at the age of two and three weeks when animals are fed by their mothers, and QTLs with highest effects after weaning when animals have to live on their own under ad libitum access to food.

Prenatal growth is very complex and a highly integrated process. Both maternal nutrition and the maternal somatotropic axis play a significant role in coordinating nutrient partitioning and utilization between maternal, placental and fetal tissues. Maternal nutrition may alter the nutrient concentrations and in turn the expression of growth regulating factors such as IGFs and IGFBPs in the blood and tissues, while GH acts in parallel via changing IGFs/IGFBPs and nutrient availability.

Myogenic cells were derived from mice long-term selected over 70 generations for high 6-weeks body weight (DU-6) and from unselected control mice (DU-Ks). The cells were grown in medium with 10% foetal bovine serum (FBS) for 8 days or transferred to low serum conditions (1% FBS) at days 4 and 6 of cultivation, respectively, and maintained for two further days. In both cell lines, serum reduction induced decreases in DNA and protein contents, and in DNA synthesis rate.

Approaches like multiple interval mapping using a multiple-QTL model for simultaneously mapping QTL can aid the identification of multiple QTL, improve the precision of estimating QTL positions and effects, and are able to identify patterns and individual elements of QTL epistasis. Because of the statistical problems in analytically deriving the standard errors and the distributional form of the estimates and because the use of resampling techniques is not feasible for several linked QTL, there is the need to perform large-scale simulation studies in order to evaluate the accuracy of multiple interval mapping for linked QTL and to assess confidence intervals based on the standard statistical theory. From our simulation study it can be concluded that in comparison with a monogenetic background a reliable and accurate estimation of QTL positions and QTL effects of multiple QTL in a linkage group requires much more information from the data.
