Enhanced Bone Remodeling After Fracture Priming.

Calcif Tissue Int

March 2022

Bone Engineering Labs, Injury, Repair & Recovery Program, Research Institute, McGill University Health Centre, 1650 Cedar Ave., Montreal, QC, H3G 1A4, Canada.

The immune system is an active component of bone repair. Mast cells influence the recruitment of macrophages, osteoclasts and blood vessels into the repair tissue. We hypothesized that if mast cells and other immune cells are sensitized to recognize broken bone, they will mount an increased response to subsequent fractures that may be translated into enhanced healing.

Circulating autoantibodies (auto-Abs) neutralizing high concentrations (10 ng/mL, in plasma diluted 1 to 10) of IFN-α and/or -ω are found in about 10% of patients with critical COVID-19 pneumonia, but not in subjects with asymptomatic infections. We detect auto-Abs neutralizing 100-fold lower, more physiological, concentrations of IFN-α and/or -ω (100 pg/mL, in 1/10 dilutions of plasma) in 13.6% of 3,595 patients with critical COVID-19, including 21% of 374 patients > 80 years, and 6.

Nowadays, no practical system has successfully been able to decode and predict pain in clinical settings. The inability of some patients to verbally express their pain creates the need for a tool that could objectively assess pain in these individuals. Neuroimaging techniques combined with machine learning are seen as possible candidates for the identification of pain biomarkers.

ICOSL in host defense at epithelial barriers: lessons from ICOSLG deficiency.

Curr Opin Immunol

October 2021

Host-directed Immunotherapy to Fight Infectious Diseases (HI-FI) Program, Research Institute - McGill University Health Centre, Montreal, Quebec, Canada; Division of Infectious Diseases, McGill University Health Centre, Montreal, Quebec, Canada; Division of Medical Microbiology, Department of Pathology & Laboratory Medicine, McGill University Health Centre, Montreal, Quebec, Canada; Department of Human Genetics, McGill University Health Centre, Montreal, Quebec, Canada. Electronic address:

Autosomal recessive mutations in Inducible T Cell Costimulator Ligand (ICOSLG) result in a combined immunodeficiency syndrome of humans, saliently marked by recurrent respiratory tract infections and significant disease with DNA-based viruses at epithelial barriers, including human papillomavirus (HPV). These features are also seen in persons with loss of function of the complementary gene, ICOS. The infection phenotypes associated with these natural experiments disclose a critical role of the corresponding proteins, ICOSL and ICOS, in human immunity at mucocutaneous barriers.

Article Synopsis
  • This study aimed to understand how patients with rheumatoid arthritis (RA) and clinicians interpret scores from PROMIS instruments used to measure pain interference and fatigue.
  • Researchers created patient scenarios to help identify symptom severity thresholds and what changes in scores are considered significant for treatment decisions.
  • Findings showed that patients typically set higher thresholds than clinicians for symptom severity and generally deemed a change of 5-10 points as meaningful, reflecting their personal experiences with RA and its impact on their daily lives.

Introduction: Radiation-based therapy (RT) has emerged as a suitable alternative to radical cystectomy (RC) and pelvic lymph node dissection for muscle-invasive bladder cancer (MIBC) patients. Routine biopsy after RT to rule out residual disease remains inconsistent across guidelines. Our objective was to review the significance of a bladder biopsy in terms of assessment of response post-RT and its potential impact on survival.

Background Hypertensive disorders of pregnancy (HDP) are associated with an increased risk of premature cardiovascular disease (CVD), but existing cardiovascular prediction models do not adequately capture risks in young women. We developed a model to predict the 10-year risk of premature CVD and mortality among women who have HDP. Methods and Results Using a population-based cohort of women with HDP who delivered between April 1989 and March 2017 in Quebec, Canada, we developed a 10-year CVD risk model using Cox proportional hazards regression.

Article Synopsis
  • Cystic fibrosis (CF) is caused by mutations in the CFTR channel, leading to issues with airflow and chronic lung inflammation due to excessive proinflammatory cytokines during infections.
  • The PDE inhibitor ensifentrine has been shown to improve CFTR function and reduce the production of inflammatory cytokines in CF bronchial cells, primarily through the inhibition of PDE4.
  • Combining ensifentrine with a beta-adrenergic agonist or corticosteroid enhances its anti-inflammatory effects, suggesting it could be a promising treatment strategy for managing inflammation in CF airways.

The neonatal fragment crystallizable (Fc) receptor (FcRn) functions as a recycling mechanism to prevent degradation and extend the half-life of IgG and albumin in the circulation. Several FcRn inhibitors selectively targeting IgG recycling are now moving rapidly toward clinical practice in neurology and hematology. These molecules accelerate the destruction of IgG, reducing pathogenic IgG and IgG immune complexes, with no anticipated effects on IgA, IgM, IgE, complement, plasma cells, B cells, or other cells of the innate or adaptive immune systems.
