Article Synopsis
  • Liquid biopsy utilizing cell-free DNA (cfDNA) analysis shows potential for non-invasive cancer diagnosis and monitoring, but the mechanisms behind cfDNA release from tumors are not fully understood.
  • A study on colorectal cancer cell lines revealed that increased cfDNA levels were linked to slower cell growth and more cell death, with less DNA methylation correlating to higher cfDNA shedding.
  • Researchers developed a methylation signature to differentiate between high and low cfDNA releasers, which was validated on additional colorectal cancer cell lines and patient-derived organoids, demonstrating its predictive capability for cfDNA release.

Skin is the forestage for a series of many-sided functions of tumor necrosis factor-alpha (TNF-α), a proinflammatory cytokine with staggering versatility and sizable implications for tissue homeostasis, immune responses, angiogenesis, apoptosis, local and systemic inflammation. An aberrant TNF-α-mediated crosstalk has been linked to the pathogenesis of acute and chronic skin inflammatory diseases, and indeed, TNF-α dysregulation can contribute to the development and progression of psoriasis, vitiligo, local damage following exposition to ultraviolet light radiations, cutaneous lupus erythematosus, and acne vulgaris. Therapies that target TNF-α are conspicuously used in the treatment of different skin disorders, aiming to modulate the in vivo immune functions triggered by many cutaneous cells, including keratinocytes, mast cells, or Langerhans cells, and reduce inflammation taking place within the skin.

Background: Breast cancer (BC) treatment has recently been revolutionized by the introduction of newer targeted agents, that helped tailoring therapies around the single patient. Along with increased survival rates, a careful evaluation of diet, lifestyle habits, physical activity, emotional and psychological experiences linked to the treatment journey, is now mandatory. However, a true proposal for an omnicomprehensive and "integrative" approach is still lacking in literature.

  • PLD-Trabectedin is an approved treatment for relapsed platinum-sensitive ovarian cancer, but its effectiveness after PARP inhibitor (PARPi) treatment hasn't been thoroughly investigated.
  • A study compared outcomes of patients who had received PARPi (cases) to those who had not (controls), finding that those previously treated with PARPi had a shorter median progression-free survival (PFS) of 8 months compared to 11 months for controls.
  • Despite the PFS difference, the tolerability of PLD-Trabectedin was similar across both groups, indicating it remains a well-tolerated treatment option even for patients who have already undergone PARPi therapy.

Predictive Factors of Response to Streptozotocin in Neuroendocrine Pancreatic Neoplasms.

J Clin Med

December 2023

Endocrinology Unit, Department of Clinical and Molecular Medicine, The European Neuroendocrine Tumor Society (ENETS) Center of Excellence, Sant'Andrea Hospital, Sapienza University of Rome, 00189 Rome, Italy.

Pancreatic neuroendocrine neoplasms (Pan-NENs) may exhibit a heterogeneous clinical course, ranging from indolent to progressive/metastatic behavior. In the latter scenario, streptozocin (STZ) is considered the cornerstone of systemic treatment; however, response to STZ-based chemotherapy may vary among individuals. In this narrative review, we aimed to identify the predictive factors of response to STZ in advanced Pan-NENs.

Int J Mol Sci

September 2023

Department of Biomedical, Surgical and Dental Sciences, University of Milan, 20122 Milan, Italy.

Extracellular vesicles (EVs) are nanosized vesicles released by almost all body tissues, representing important mediators of cellular communication, and are thus promising candidate biomarkers for neurodegenerative diseases like Alzheimer's disease (AD). The aim of the present study was to isolate total EVs from plasma and characterize their microRNA (miRNA) contents in AD patients. We isolated total EVs from the plasma of all recruited subjects using ExoQuickULTRA exosome precipitation solution (SBI).
