Mitochondrial malic enzyme 2 (ME2), which catalyzes the conversion of malate to pyruvate, is frequently upregulated during tumorigenesis and is a potential target for cancer therapy. However, the regulatory mechanism underlying ME2 activity is largely unknown. In this study, we demonstrate that ME2 is highly expressed in human colorectal cancer (CRC) tissues, and that ME2 knockdown inhibits the proliferation of CRC cells.

We have studied the stability of the smallest long-lived all carbon molecular dianion (C_{7}^{2-}) in new time domains and with a single ion at a time using a cryogenic electrostatic ion-beam storage ring. We observe spontaneous electron emission from internally excited dianions on millisecond timescales and monitor the survival of single colder C_{7}^{2-} molecules on much longer timescales. We find that their intrinsic lifetime exceeds several minutes-6 orders of magnitude longer than established from earlier experiments on C_{7}^{2-}.

Folate-mediated one-carbon metabolism (FOCM) is crucial in sustaining rapid proliferation and survival of cancer cells. The folate cycle depends on a series of key cellular enzymes, including aldehyde dehydrogenase 1 family member L2 (ALDH1L2) that is usually overexpressed in cancer cells, but the regulatory mechanism of ALDH1L2 remains undefined. In this study, we observed the significant overexpression of ALDH1L2 in colorectal cancer (CRC) tissues, which is associated with poor prognosis.

Systemic acquired resistance (SAR) is a potent innate immunity system in plants that is induced through the salicylic acid (SA)-mediated signaling pathway. Here, we characterized 3-chloro-1-methyl-1-pyrazole-5-carboxylic acid (CMPA) as an effective SAR inducer in . The soil drench application of CMPA enhanced a broad range of disease resistance against the bacterial pathogen and fungal pathogens and in , whereas CMPA did not show antibacterial activity.

Idiopathic pulmonary fibrosis (IPF) is a progressive fibrosing lung disease that is caused by the dysregulation of alveolar epithelial type II cells (AEC II). Proton-dependent oligopeptide transporters (POTs) belong to the major facilitator superfamily (MFS) and transport dipeptides and tripeptides from the extracellular environment into the target cell. The human POTs PepT1 and PepT2 are also involved in the absorption of various orally ingested drugs. Previously reported structures revealed that the bacterial POTs possess 14 helices, of which H1-H6 and H7-H12 constitute the typical MFS fold and the residual two helices are involved in the cytoplasmic linker.

ERECT LEAF 1 (ELF1), which was identified as a component of brassinosteroid signaling in rice, is involved in brassinosteroid-mediated suppression of jasmonic acid response. Here, by conducting yeast two-hybrid assay and in vitro ubiquitination experiments, we demonstrate that ELF1 interacts with the OsWRKY4 transcription factor, a positive regulator of defense responses to rice sheath blight. ELF1 decreased the stability of OsWRKY4, whereas exogenous jasmonic acid treatment suppressed this effect of ELF1, resulting in OsWRKY4 accumulation in rice plants.

A novel biodegradable conducting polymer, PLA-b-AP-b-PLA (PAP) triblock copolymer of poly (l-lactide) (PLA) and aniline pentamer (AP) with electroactivity and biodegradability, was synthesized and its potential application in cardiac tissue engineering was studied. The PAP copolymer presented better biocompatibility compared to PANi and PLA because of promoted cell adhesion and spreading of rat cardiac myoblasts (H9c2 cell line) on PAP/PLA thin film. After pulse electrical stimulation (5 V, 1 Hz, 500 ms) for 6 days, the proliferation ratio, and intracellular calcium concentration of H9c2 cells on PAP/PLA were improved significantly.

DNA double-strand break (DSB)-mediated genome rearrangements are assumed to provide diverse raw genetic materials enabling accelerated adaptive evolution; however, it remains unclear about the consequences of massive simultaneous DSB formation in cells and their resulting phenotypic impact. Here, we establish an artificial genome-restructuring technology by conditionally introducing multiple genomic DSBs in vivo using a temperature-dependent endonuclease TaqI. Application in yeast and Arabidopsis thaliana generates strains with phenotypes, including improved ethanol production from xylose at higher temperature and increased plant biomass, that are stably inherited to offspring after multiple passages.

Histone H3 lysine 9 (H3K9) methylation is unevenly distributed in mammalian chromosomes. However, the molecular mechanism controlling the uneven distribution and its biological significance remain to be elucidated. Here, we show that JMJD1A and JMJD1B preferentially target H3K9 demethylation of gene-dense regions of chromosomes, thereby establishing an H3K9 hypomethylation state in euchromatin.

The first free cyclobutadiene (CBD) germanium analogue was obtained as room-temperature stable dark red crystals via the reaction of the bulky EMind-substituted 1,2-dichlorodigermene with lithium naphthalenide. The cyclic 4π-electron antiaromaticity is essentially stabilized by the polar Jahn-Teller distortion in the germanium CBD producing a planar rhombic-shaped charge-separated structure.

Symbiotic protists in the hindgut of termites provide a novel enzymatic resource for efficient lignocellulytic degradation of plant biomass. In this study, two β-mannanases, RsMan26A and RsMan26B, from a symbiotic protist community of the lower termite, Reticulitermes speratus, were successfully expressed in the methylotrophic yeast, Pichia pastoris. Biochemical characterization experiments demonstrated that both RsMan26A and RsMan26B are endo-acting enzymes and have a very similar substrate specificity, displaying a higher catalytic efficiency to galactomannan from locust bean gum (LBG) and glucomannan than to β-1,4-mannan and highly substituted galactomannan from guar gum.

ERECT LEAF 1 (ELF1) was previously identified as a component of brassinosteroid signaling in rice. A double mutant obtained by crossing elf1-1 (a null mutant of ELF1) with d61-1 (a leaky mutant of OsBRI1) showed a more severe phenotype than did the elf1-1 single mutant, resembling that of a severe brassinosteroid-deficient mutant. Microarray analysis showed that the gene expression profile of elf1-1 was distinct from that of d61-12 (a leaky mutant of OsBRI1 with a phenotype similar to that of elf1-1), and fewer than half of genes differentially expressed between the wild-type and elf1-1 showed similar differences in d61-12 relative to the wild-type.

We have investigated the terahertz-spectral property of nicotinamide focusing on the temperature dependence in the range of 14-300 K. We observed that almost all peaks in the terahertz spectrum of the nicotinamide crystal showed a remarkable shift with temperature, whereas the lowest-frequency peak at 34.8 cm showed a negligible shift with temperature.

How to orient polymers homeotropically in thin films has been a long-standing issue in polymer science because polymers intrinsically prefer to lie down. Here we provide a design principle for polymers that are processable into a 2D homeotropic order. The key to this achievement was a recognition that cylindrical polymers can be designed to possess oppositely directed local dipoles in their cross-section, which possibly force polymers to tightly connect bilaterally, affording a 2D rectangular assembly.
