Host-dependent C-to-U RNA editing in SARS-CoV-2 creates novel viral genes with optimized expressibility.

Front Cell Infect Microbiol

October 2024

Qingdao Hospital of Traditional Chinese Medicine, Qingdao Haici Hospital, Qingdao, Shandong, China.

Rampant C-to-U RNA editing drives the mutation and evolution of SARS-CoV-2. While much attention has been paid to missense mutations, the C-to-U events leading to AUG and thus creating novel ORFs were uninvestigated. By utilizing the public time-course mutation data from the worldwide SARS-CoV-2 population, we systematically identified the "AUG-gain mutations" caused by C-to-U RNA editing.

Background: Cognitive impairments are a hallmark symptom of schizophrenia (SCZ). Phosphatidylethanolamine (PE) is the second most abundant phospholipid in mammalian cells, yet its role in cognitive deficits remains unexplored. The aim of this study was to investigate the association between plasma LysoPE and cognitive improvements following olanzapine monotherapy in drug-naïve first-episode (DNFE) SCZ patients.

Background: It has been established that there are functional changes in the brain of treatment-resistant depression (TRD) patients, but previous studies of functional connectivity (FC) usually involved selection of regions of interest based on accumulated a priori knowledge of the disorder. In this study, we combine amplitude of low-frequency fluctuation (ALFF) and FC; this approach, based on the abnormal ALFF, may provide some insights into the neural basis of the disease in terms of fMRI signals of low-frequency fluctuations.

Methods: A total of 16 TRD patients, who visited the Qingdao Mental Health Center, Shandong Province, China between March 2023 and January 2024, along with 16 normal subjects, were enrolled into this study for functional imaging.

  • The study aims to analyze the relationship between schizophrenia and the risk of developing type 2 diabetes mellitus (T2DM) by integrating recent observational studies.
  • A comprehensive literature search was performed, and various statistical methods were used to evaluate the studies, including the assessment of bias and heterogeneity.
  • Results from 32 studies indicated a significant association between schizophrenia and a higher risk of T2DM, particularly showing that females with schizophrenia have an even greater risk compared to their male counterparts.

  • 5-fluorouracil (5-Fu) is an effective chemotherapy drug, but its use is limited due to harmful effects on the heart.
  • Natural compounds like resveratrol (RES) may help counteract these cardiotoxic effects while boosting the anti-cancer benefits of 5-Fu.
  • In tests, combining RES with 5-Fu not only reduced tumor growth and improved cancer cell responses but also protected heart cells from damage, making RES a promising adjunct therapy for gastric cancer treatment.

Article Synopsis
  • A diagnostic model for Bipolar Disorder (BD) during its depressive phase was developed using RNA data from patients, employing a combination of Random Forest and Feedforward Neural Networks for analysis.
  • The study utilized multiple datasets to identify key differentially expressed genes and trained a neural network model while implementing techniques to avoid overfitting.
  • The final model exhibited strong performance metrics, with a specificity of 0.769, sensitivity of 0.818, and an AUC of 0.832, concluding that this approach effectively classifies BD based on significant gene expression changes.

Neutrophil/lymphocyte ratio and cognitive performances in first-episode patients with schizophrenia and healthy controls.

Prog Neuropsychopharmacol Biol Psychiatry

December 2024

Peking University HuiLongGuan Clinical Medical School, Beijing HuiLongGuan Hospital, Beijing, China. Electronic address:

Abnormal immune and inflammatory responses are considered to contribute to schizophrenia (SZ). The neutrophil/lymphocyte ratio (NLR) is an inexpensive and reproducible marker of systemic inflammatory responses. Accumulating studies have demonstrated that NLR values are increased in SZ compared to healthy controls and closely related to clinical symptoms in antipsychotic-naïve first-episode SZ (ANFES) patients.
