Cancer-associated Fibroblasts (CAFs) have emerged as critical regulators of anti-tumour immunity, with both beneficial and detrimental properties that remain poorly characterised. To investigate this, we performed single-cell and spatial transcriptomic analysis, comparing head & neck squamous cell carcinoma (HNSCC) subgroups, which although heterogenous, can be considered broadly immune-hot and immune-cold (human papillomavirus [HPV]+ve and HPV-ve tumours respectively). This identified six fibroblast subpopulations, including two with immunomodulatory gene expression profiles (IL-11 + inflammatory [i]CAF and CCL19 + fibroblastic reticular cell [FRC]-like).

Hybrid architecture based intelligent diagnosis assistant for GP.

BMC Med Inform Decis Mak

January 2024

National Centre for Computer Animation, Bournemouth University, Bournemouth, UK.

As the first point of contact for patients, General Practitioners (GPs) play a crucial role in the National Health Service (NHS). An accurate primary diagnosis from the GP can alleviate the burden on specialists and reduce the time needed to re-confirm the patient's condition, allowing for more efficient further examinations. However, GPs have broad but less specialized knowledge, which limits the accuracy of their diagnosis.

Article Synopsis
  • The study aimed to analyze the effects of diabetes on cardiac remodelling and exercise capacity in patients with aortic stenosis (AS).
  • It found that while diabetes patients had similar heart volumes and function compared to non-diabetic patients, they exhibited worse exercise capacity and higher levels of certain biomarkers.
  • Importantly, diabetes did not significantly increase cardiovascular events, but it was linked to a higher risk of all-cause mortality, highlighting its need for careful management in AS patients.

Cerebral blood flow (CBF) is an important physiological parameter that can be quantified non-invasively using arterial spin labelling (ASL) imaging. Although most ASL studies are based on single-timepoint strategies, multi-timepoint approaches (multiple-PLD) in combination with appropriate model fitting strategies may be beneficial not only to improve CBF quantification but also to retrieve other physiological information of interest. In this work, we tested several kinetic models for the fitting of multiple-PLD pCASL data in a group of 10 healthy subjects.
