  • Mevalonate kinase deficiency is a rare genetic disorder caused by mutations in the MVK gene, leading to various autoinflammatory diseases, including hyper-IgD syndrome and mevalonic aciduria.
  • Approximately 300 cases have been documented globally, showcasing its rarity.
  • A 3-year-old girl from Puerto Rico was diagnosed with hyper-IgD syndrome after experiencing fever, joint pain, and skin lesions from an early age; she will be treated with canakinumab following routine vaccinations.

The comorbidity between cocaine use disorder (CUD) and trauma/stressor-related disorders suggests a connection between neurophysiological changes induced by stress and those that lead to cocaine use. Due to the unexpected and sometimes uncontrollable nature and timing of stressful life events, their capacity to induce drug use poses a significant challenge for the administration of cocaine relapse therapy. This study aims to investigate the impact of chronic stress applied prior to cocaine acquisition on the development of cocaine-seeking behavior after different periods of drug abstinence in male and female rats.

Arachnoid granulations (AGs) are generally benign structures within the subarachnoid space that extend into the dural sinuses and calvarial bone. They can present in a variety of sizes but are termed 'giant' arachnoid granulations (GAGs) when they are larger than 1 cm in diameter or take up a significant portion of the dural sinus' lumen. Vermiform giant arachnoid granulations are a specific type of GAG that are known for their worm-like appearance.

Currently, there is limited data available comparing Primary Mediastinal Large B-cell Lymphoma (PMBL) and mediastinal Hodgkin disease, nodular sclerosis type (HDNS). This is a retrospective cohort study that compares the clinical features, histology through immunohistochemistry (IHC) and treatment outcomes of 19 cases of PMBL and 39 cases of HDNS diagnosed over 13 years at a single institution in San Juan, PR. Superior Vena Cava syndrome (SVCS) and elevated Lactate Dehydrogenase (LDH) levels were more frequently seen in the PMBL cohort.

GABA system as the cause and effect in early development.

Neurosci Biobehav Rev

June 2024

Division of Basic Neuroscience, McLean Hospital, Belmont, MA 02478, USA; Department of Psychiatry, Harvard Medical School, Boston, MA 02215, USA. Electronic address:

GABA is the primary inhibitory neurotransmitter in the adult brain and through its actions on GABARs, it protects against excitotoxicity and seizure activity, ensures temporal fidelity of neurotransmission, and regulates concerted rhythmic activity of neuronal populations. In the developing brain, the development of GABAergic neurons precedes that of glutamatergic neurons and the GABA system serves as a guide and framework for the development of other brain systems. Despite this early start, the maturation of the GABA system also continues well into the early postnatal period.

Importance: Strategies to reduce medication dosing errors are crucial for improving outcomes. The Medication Education for Dosing Safety (MEDS) intervention, consisting of a simplified handout, dosing syringe, dose demonstration and teach-back, was shown to be effective in the emergency department (ED), but optimal intervention strategies to move it into clinical practice remain to be described.

Objective: To describe implementation of MEDS in routine clinical practice and associated outcomes.

Laser-Assisted Prostaglandin Analogs in the Treatment of Hypopigmented Scars: A Systematic Review.

J Clin Aesthet Dermatol

September 2023

Drs. Lal and Goldberg are with the Skin Laser and Surgery Specialists Division of Schweiger Dermatology in New York, New York.

Background: Hypopigmented scars are challenging to treat due to a lack of effective treatments and often transient results. Recent reports suggest that prostaglandin analog-induced hyperpigmentation may have favorable dermatological applications.

Objective: Analyze previous studies involving the use of prostaglandin analogs in the treatment of hypopigmented scars.

Article Synopsis
  • The US faces a significant gun violence issue, highlighted by 698 mass shootings in 2021, resulting in 705 deaths and thousands of injuries.
  • Researchers analyzed clinical data from 31 hospitals regarding 403 survivors of mass shootings from 2012-2019, focusing on both physical and psychological injuries.
  • Findings revealed that most victims sustained serious physical injuries (primarily from gunshot wounds) and many experienced psychiatric issues, indicating the severe impact of mass shootings on survivors' health and well-being.

Article Synopsis
  • The study investigates the differences between adult-onset atopic dermatitis (AOAD) and pediatric-onset atopic dermatitis persisting into adulthood (POAD), focusing on their unique characteristics to aid in targeted therapy development.
  • Skin biopsies and blood samples from adults with AOAD, POAD, and healthy controls were analyzed using various techniques, revealing distinct immune responses and dysregulations in both conditions.
  • Results indicated that while POAD showed more severe inflammation and epidermal barrier issues, AOAD had different immune marker expressions and a greater number of dysregulated proteins in serum, suggesting the need for diverse therapeutic strategies that go beyond just targeting Th2 pathways.

(1) Purpose: A patient with scleritis may have an associated systemic disease, which is often autoimmunological and seldom infectious in origin. The data regarding such associations in Hispanic populations are scarce. Therefore, we evaluated the clinical characteristics and systemic-disease associations of a cohort of Hispanic patients with scleritis.

Objective: Examine whether DNA repair capacity (DRC) levels are associated with body mass index (BMI) in adult women.

Design And Participants: A nested study composed of 539 women without breast cancer (BC) from a case-control BC study in addition to 104 that were recruited later for a total of 643.

Measurements: DRC levels were measured in lymphocytes using a host-cell reactivation assay with a luciferase reporter gene damaged by UVC.

Background: Brown adipose tissue (BAT) plays a role in modulating energy expenditure. People with obesity have been shown to have reduced activation of BAT. Agents such as β-agonists, capsinoids, thyroid hormone, sildenafil, caffeine, or cold exposure may lead to activation of BAT in humans, potentially modulating metabolism to promote weight loss.

Article Synopsis
  • Janus kinase (JAK) inhibitors, specifically ritlecitinib and brepocitinib, are being studied for treating alopecia areata (AA) with promising results from early research.
  • In a phase 2a clinical trial, biopsy samples from 46 patients showed significant improvements in scalp biomarkers after 24 weeks, correlating with hair regrowth measured by the Severity of Alopecia Tool (SALT) score.
  • While brepocitinib showed better results at week 12, ritlecitinib outperformed it by week 24, suggesting the need for larger and longer-term trials to confirm these findings.
