Article Synopsis
  • Protein lactylation is a common post-translational modification found in proteins, particularly histones, and plays a role in the initiation and progression of tumors, including triple-negative breast cancer (TNBC).
  • In a study involving 77 TNBC tissue samples, researchers found that while pan-lysine lactylation (panKlac) levels were higher in tumor tissues compared to normal areas, they did not correlate with traditional clinical factors like tumor size or lymph node involvement.
  • High levels of nuclear panKlac were linked to better recurrence-free survival for patients, while elevated cytoplasmic panKlac appeared to protect against poor outcomes; further analysis suggested that lactylation influences pathways related to cell growth and might involve specific

Objective: Schizophrenia belongs to a severe mental illness with complicated clinical presentations, an ill-defined pathogenesis, and no known cause. Many genetic studies imply that polygenic interaction is important in the development of schizophrenia. The main mechanism of the RELN-BDNF-CREB-DNMT signaling pathway in neurodevelopment involves RELN, brain-derived neurotrophic factor (BDNF), transcription factor cyclic adenosine monophosphate response element binding protein (CREB), DNA methyltransferase 1 (DNMT1), as well as DNA methyltransferase 3B (DNMT3B).

Article Synopsis
  • - Checkpoint inhibitors like tislelizumab have shown effectiveness in treating recurrent/metastatic nasopharyngeal cancer (R/M NPC), significantly improving progression-free survival (PFS) compared to placebo when combined with chemotherapy.
  • - In a clinical trial (RATIONALE-309), patients receiving tislelizumab with chemotherapy had better outcomes regardless of their programmed death-ligand 1 expression, and the overall safety of the treatment was similar to the placebo group.
  • - Gene expression profiling identified specific tumor characteristics that were associated with better PFS in patients treated with tislelizumab-chemotherapy, suggesting this combination should be considered for first-line treatment in R/M NPC and could help select the best candidates for therapy

Background: The causal inference between leisure sedentary behaviour (LSB) and rheumatoid arthritis (RA) is still controversial because of potential residual confounding and reverse causality.

Methods: The present study used publicly available large-scale genome-wide association studies (GWAS) of LSB (television watching, computer use, and driving) and RA to perform a two-sample Mendelian randomization (MR) study to evaluate the causal effect of LSB on the risk of RA. We detected significant causal associations using the multiplicative random effects-inverse variance weighted (MRE-IVW) method, the maximum likelihood, robust adjusted profile scores, the weighted median, MR-Egger regression, and several complementary sensitivity analyses.
