Article Synopsis
  • Scientists are looking at how confidence scores help check if a computer program called AlphaFold (AF2) is good at predicting the shapes of proteins, which is important for making new medicines.!
  • They found that ignoring low confidence scores (below 80) helps them get better predictions because those scores can be messy due to some proteins being disordered.!
  • Out of the latest crystal structures they studied, 95% probably have the correct shapes, and AF2 could help find useful info about many human proteins that are not well understood yet.!

Three-dimensional (3D) cell culture models are widely used in tumor studies to more accurately reflect cell-cell interactions and tumor growth conditions . 3D anchorage-independent spheroids derived by culturing cells in ultra-low attachment (ULA) conditions is particularly relevant to ovarian cancer, as such cell clusters are often observed in malignant ascites of late-stage ovarian cancer patients. We and others have found that cells derived from anchorage-independent spheroids vary widely in gene expression profiles, proliferative state, and metabolism compared to cells maintained under attached culture conditions.

Background: The objective of this study was to test the hypothesis that exercise would be more effective than a support group plus Fitbit (SG+Fitbit) program in improving functional outcomes in older breast cancer survivors (BCSs) and that race would moderate the exercise effect on outcomes.

Methods: Older African American (AA) and non-Hispanic White (NHW) BCSs were purposively recruited and enrolled into the 52-week randomized controlled trial. The interventions included 20 weeks of supervised moderate-intensity aerobic and resistance training followed by 32 weeks of unsupervised exercise called IMPROVE (n = 108) and a 20-week SG+Fitbit program followed by 32 weeks of unsupervised activity (n = 105).

Objectives: Insomnia symptoms are transdiagnostic to physical and mental health disorders. Given the lack of population-based cohorts with objective sleep measures and long-term follow-ups, little is known about the chronicity of childhood insomnia symptoms. We determined the developmental trajectories of insomnia symptoms, their evolution into adult insomnia, and the role of objective sleep duration in the transition to adulthood.

Article Synopsis
  • Cardiac resynchronization therapy (CRT) shows potential benefits in patients with permanent atrial fibrillation (AF) but limited data exists on its effectiveness, leading to the investigation of atrio-ventricular junctional ablation (AVJA).
  • A study involving 26 patients randomized to receive CRT-D with or without AVJA found no significant difference in left ventricular end-systolic volume (LVESV) improvement between the two groups at 6 months.
  • Although AVJA didn't appear to enhance outcomes, CRT demonstrated effectiveness overall in improving left ventricular ejection fraction (LVEF) and functional class among patients with AF.

Background: In this post hoc analysis, we assessed patient characteristics as predictors of dronedarone trough concentrations and characterized the relationship of trough concentrations of dronedarone with its efficacy and safety.

Hypothesis: Dronedarone is recommended as a 400 mg twice daily dose taken orally with meals. We hypothesize that drug concentration/bioavailability of dronedarone, measured as above- and below-median trough concentrations, does not impact the efficacy outcomes.

Article Synopsis
  • Novel mRNA vaccines were first distributed in the U.S. to high-risk individuals, including smokers, starting in December 2020, with over half of the adult population receiving at least one dose by early 2021.
  • A study of 231 adult smokers in Pennsylvania in April 2021 found that nearly 60% had received at least one vaccine dose, but 84% of the unvaccinated reported they were unlikely to get vaccinated.
  • Vaccine hesitancy was linked to lower consumption of COVID-19 news and distrust in government information, with concerns about vaccine safety and side effects being common among those hesitant to vaccinate.

Development is a sophisticated process maintained by various signal transduction pathways, including the Hedgehog (Hh) pathway. Several important functions are executed by the Hh signaling cascade such as organogenesis, tissue regeneration, and tissue homeostasis, among various others. Considering the multiple functions carried out by this pathway, any mutation causing aberrant Hh signaling may lead to myriad developmental abnormalities besides cancers.

Deficits in neuronal structure are consistently associated with neurodevelopmental illnesses such as autism and schizophrenia. Nonetheless, the inability to access neurons from clinical patients has limited the study of early neurostructural changes directly in patients' cells. This obstacle has been circumvented by differentiating stem cells into neurons, although the most used methodologies are time consuming.

Research progress on dopamine D receptors indicates that signaling no longer is limited to G protein-dependent cyclic adenosine monophosphate phosphorylation but also includes G protein-independent β-arrestin-related mitogen-activated protein kinase activation, regulation of ion channels, phospholipase C activation, and possibly more. This review summarizes recent studies revealing the complexity of D signaling and its clinical implications, and suggests functional selectivity as a promising strategy for drug discovery to magnify the merit of D signaling. Functional selectivity/biased receptor signaling has become a major research front because of its potential to improve therapeutics through precise targeting.
