A 50-year-old man was admitted to the hospital with a chief complaint of long-standing cold-induced numbness in the fingers for more than a year, accompanied by persistent hand pain and fingertip ulceration for 3 months. On physical examination, radial and ulnar artery pulsation was absent in both limbs, and pale skin color, low skin temperature, and limited finger range of motion were noted. Gangrene was detected in the fingertips of the left index and middle fingers, and the right middle and ring fingers.

College of Biomedical Engineering, National Engineering Research Center for Biomaterials, Sichuan University, Chengdu, 610064, China.

Near-infrared (NIR) light, compared with ultraviolet (UV) light, has a stronger tissue penetration ability and is widely used in the medical field. However, few hydrogels can be triggered by NIR. Here, a modular polymer-nanosheet (metal disulfide) (PNS) hydrogel system was proposed, which can be photo-crosslinked through photothermal conversion under NIR light.

Acute lung injury (ALI) is a serious disorder characterized by the release of pro-inflammatory cytokines and cascade activation of macrophages. Ferroptosis, a form of iron-dependent cell death triggered by intracellular phospholipid peroxidation, has been implicated as an internal mechanism underlying ALI. In this study, we investigated the effects of mA demethylase fat mass and obesity-associated protein (FTO) on the inhibition of macrophage ferroptosis in ALI.

Cisplatin (DDP) based combination chemotherapy is a vital method for the treatment of bladder cancer (BLca). Chemoresistance easily occurs in the course of cisplatin chemotherapy, which is one of the important reasons for the unfavorable prognosis of BLca patients. Circular RNAs (circRNAs) are widely recognized for their role in the development and advancement of BLca.

YTH domain family protein 3 accelerates non-small cell lung cancer immune evasion through targeting CD8 T lymphocytes.

Cell Death Discov

July 2024

Department of Thoracic Surgery, The First Affiliated Hospital of Shenzhen University, Shenzhen Second People's Hospital, Shenzhen, 518000, Guangdong Province, China.

Article Synopsis
  • Scientists learned that a protein called YTHDF3 helps cancer cells in non-small cell lung cancer (NSCLC) avoid attacks from the immune system.
  • When YTHDF3 levels are high, it stops CD8 T cells (a type of immune cell) from fighting the cancer effectively.
  • This study shows that targeting YTHDF3 may help boost the immune system's ability to fight lung cancer, providing ideas for new treatments.

Cell Death Discov

July 2024

Shenzhen Key Laboratory of Immunity and Inflammatory Diseases, Peking University Shenzhen Hospital, Shenzhen, China.

Cancer immunotherapy harnesses the body's immune system to combat malignancies, building upon an understanding of tumor immunosurveillance and immune evasion mechanisms. This therapeutic approach reactivates anti-tumor immune responses and can be categorized into active, passive, and combined immunization strategies. Active immunotherapy engages the immune system to recognize and attack tumor cells by leveraging host immunity with cytokine supplementation or vaccination.

Electronic address:

Article Synopsis
  • - The study investigates why some patients don’t benefit from PD-1/PD-L1 cancer therapies, focusing on the intrinsic roles of PD-L1 in head and neck squamous cell carcinoma (HNSCC) cells.
  • - Using a CRISPR screen, researchers found that PD-L1-deficient cells rely on certain ferroptosis-related genes for survival and are more prone to cell death when ferroptosis is induced.
  • - PD-L1 appears to protect HNSCC cells from ferroptosis by boosting the antioxidant SOD2, which keeps reactive oxygen species (ROS) levels low, suggesting that targeting PD-L1's functions could improve treatment outcomes.

Article Synopsis
  • - The study investigates the effects of ASC22 (a PD-1 antibody) on patients with chronic hepatitis B who are already virally suppressed using nucleos(t)ide analogs, aiming to find a potential cure for HBV by targeting the PD-1 pathway.
  • - In a phase IIb trial involving 149 patients, those receiving 1.0 mg/kg ASC22 experienced significant declines in HBsAg levels compared to the placebo group, with 30% achieving HBsAg loss among those with lower baseline levels.
  • - The treatment was found to be safe and well-tolerated, with mostly mild adverse events reported, indicating ASC22 has potential as a therapeutic option for chronic hepatitis B infection.

Tumor-stromal interactions and stromal heterogeneity in the tumor microenvironment are critical factors that influence the progression, metastasis, and chemoresistance of pancreatic ductal adenocarcinoma (PDAC). Here, we used spatial transcriptome technology to profile the gene expression landscape of primary PDAC and liver metastatic PDAC after bioactive black phosphorus nanomaterial (bioactive BP) treatment using a murine model of PDAC (; ; and mice). Bioinformatic and biochemical analyses showed that bioactive BP contributes to the tumor-stromal interplay by suppressing cancer-associated fibroblast (CAF) activation.

Fbxo28 is essential for spindle migration and morphology during mouse oocyte meiosis I.

Int J Biol Macromol

August 2024

Department of Obstetrics and Gynecology, Tangdu Hospital, Air Force Medical University, Xi'an 710038, China. Electronic address:

Article Synopsis
  • Fbxo28 is an important protein expressed in oocytes that plays a crucial role in spindle migration and assembly during asymmetric oocyte division, which is vital for fertility.
  • The study utilized various techniques, including microinjection and microscopy, to investigate the function of Fbxo28, revealing that it primarily localizes to chromosomes and microtubule-organizing centers.
  • Results showed that knocking down Fbxo28 led to defects in spindle shape and movement without affecting polar body extrusion, and these issues could be reversed by supplementing with Fbxo28-myc mRNA.
