Purpose: Waldenström macroglobulinemia (WM) is a rare type of lymphoma, with no optimal treatment. BTK inhibitor have shown promising outcomes, yet achieving deep remission (VGPR or CR) remains challenging and time-limited therapy with proteasome-inhibition has not been reported. We conducted a phase 2 clinical trial (NCT04463953) to evaluate the efficacy and safety of combining zanubrutinib, ixazomib and dexamethasone (ZID) in newly diagnosed WM patients.

Background: The aim of this study was to evaluate the efficacy of doxycycline-sitafloxacin sequential therapy in the treatment of Mycoplasma genitalium (Mg) urogenital infections in Nanjing, China.
Methods: Potential subjects were tested initially for Mg infection by nucleic acid amplification testing and again at least 21 days after completion of doxycycline (100 mg twice daily for 7 days)-sitafloxacin (100 mg twice daily for 7 days) sequential therapy. The presence of macrolide and quinolone resistance-associated mutations in 23S rRNA, parC, gyrA, and gyrB genes in Mg was examined at baseline and upon retesting of specimens from subjects that did not clear Mg.

Purpose: Brain structure-function coupling (SFC), which reflects the degree to which anatomical structure supports neural function, is an emerging imaging marker in neurodegenerative diseases. However, its pathological underpinnings in Alzheimer's disease (AD) remain poorly understood. This study aimed to examine the association among amyloid pathology, SFC disruption and cognitive decline.

Objectives: To improve the accuracy of lymphocyte subsets counts, a traceable dual-platform extended PanLeucogating method (DPP) of absolute cell counts of lymphocyte subsets was introduced, and consistency was evaluated by comparing conventional single/dual-platform method with DPP.
Methods: The DPP for absolute lymphocyte subsets counts was established by multiplying the percentage of lymphocyte subsets in total white blood cells (WBC) measured by flow cytometer with the total WBC counts. DPP-R was defined as the use of the total WBC counts measured in reference laboratory that was traceable to reference method recommended by International Council for Standardization in Hematology (ICSH).

Glioblastoma multiforme (GBM) is an aggressive and lethal primary brain tumor with limitedtreatment options due to its resistance to conventional therapies and an immunosuppressive tumor microenvironment. In this study, we investigated whether clofazimine, an inhibitor of the Wnt/β-catenin signaling pathway, could enhance the efficacy of anti-PD-1 immunotherapy in GBM. Our in vitro and in vivo experiments demonstrated that clofazimine suppressed GBM cell proliferation, induced apoptosis, and inhibited invasion by downregulating Wnt6-mediated activation of the Wnt/β-catenin pathway and the downstream MEK/ERK signaling cascade, leading to decreased PD-L1 expression.

Coptisine (COP), a naturally occurring alkaloid, is recognized for its varied pharmacological impacts and its supportive function in intestinal well-being. However, the role of COP to protect the colonic epithelium in colitis has not been extensively investigated. The objective of this study was to assess the efficacy of COP in ameliorating colitis by investigating intestinal histopathology, mucosal barrier function, and transient receptor potential (TRP) signaling pathways in mice with colon disease compared to a control group, thereby elucidating the underlying mechanisms of its action.

Intervertebral disc (IVD) degeneration (IVDD), primarily caused by nucleus pulposus (NP) dehydration, leads to low back pain. While current treatments focus on symptom management or surgical intervention, tissue engineering using IVD-derived cells, biofactors, and scaffolds offers a promising regenerative approach. Here, human NP cells (NPCs) and annulus fibrosus cells (AFCs) are immortalized with human telomerase reverse transcriptase (hTERT), generating immortalized NPCs (iHNPCs) and AFCs (iHAFCs).

Purpose: To evaluate choroidal changes in high myopic (HM) patients with posterior staphyloma (PS) and explore their association with Gaussian curvature (K) using ultra-wide-field optical coherence tomography angiography (UWF-OCTA).
Methods: HM patients with wide macular PS (Type I PS) and without any PS (control) were recruited. Highest K values (K) and choroidal parameters-vessel flow density (VFD), choroidal vascularity volume (CVV) and index (CVI), thickness, and volume-were compared in central and peripheral regions, and the fovea-centered 12-mm ETDRS quadrants of 24 × 20 mm UWF-OCTA images.

Background: Dual antiplatelet therapy (DAPT) with clopidogrel and aspirin is recommended for secondary prevention in patients with a minor stroke or transient ischemic attack. However, the effectiveness of DAPT can be significantly influenced by genetic variations. This study aimed to estimate the impact of multiple single-nucleotide polymorphisms across various genes on DAPT efficacy using polygenic risk score (PRS).

Background: Venous thromboembolism is a significant complication after knee replacement. The short-term efficacy disparities between different types of graduated elastic compression stockings (GCS) among patients undergoing total/unicompartmental knee replacement remain unclear.
Objective: The aim of the trial was to compare the efficacies on hemodynamics and morphology of femoral vein between two types of GCS, providing more evidence on GCS prophylaxis among patients undergoing total/unicompartmental knee replacement.

Capillary hemangiomas are commonly encountered superficially in the cutaneous, subcutaneous, or mucosal tissues during childhood and early adulthood, but retroperitoneal capillary hemangioma is a rare occurrence. We present a case of a 61-year-old woman with incidentally detected capillary hemangioma in the retroperitoneum initially presumed to be paraganglioma based on 18F-AlF-NOTA-Octreotide PET/CT findings. Contrast-enhanced MRI revealed a hypervascular mass in the retroperitoneum.

Background: Lack of data integrity is a common problem in randomized clinical trials and is more serious in economic evaluations conducted alongside explanatory clinical trials. Despite pragmatic randomized controlled trials (pRCTs) becoming recognized as the best design for economic evaluations, information on the proportion, handling approaches, and reporting quality of missing data in pRCTs-based economic evaluations remains limited. This study aimed to investigate the quantity and reporting quality of missing data in economic evaluations conducted alongside pragmatic clinical trials.

Whole-body bone scan (WBS) is usually used as the effective diagnostic method for early-stage and comprehensive bone metastases of breast cancer. WBS images with breast cancer bone metastasis have the characteristics of low resolution, small foreground, and multiple lesions, hindering the widespread application of deep learning-based models. Automatically detecting a large number of densely small lesions on low-resolution WBS images remains a challenge.

Diabetic retinopathy (DR) is one of the most common microvascular complications of diabetes mellitus and a leading cause of vision loss in diabetic patients. Pathological processes such as inflammatory response, oxidative stress, and vascular dysfunction play important roles in the pathogenesis of DR. Changes in the levels of cytokines associated with these pathologic processes correlate with the onset, progression, and extent of DR.
