Huntington's disease (HD) is a debilitating neurodegenerative disorder characterized by severe motor deficits, cognitive decline and psychiatric disturbances. An early and significant morphological hallmark of HD is the activation of astrocytes triggered by mutant huntingtin, leading to the release of inflammatory mediators. Fingolimod (FTY), an FDA-approved sphingosine-1-phosphate (S1P) receptor agonist is used to treat multiple sclerosis (MS), a neuroinflammatory disease, and has shown therapeutic promise in other neurological conditions.

Arbuscular mycorrhiza (AM) represents a symbiotic mutualistic association between most land plants and fungi. AM fungi develops specialized intraradical and highly branched structures, called arbuscules, where bidirectional exchange of nutrients between plant and fungi partners occurs, improving plant growth and fitness. Transcriptional reprogramming and hormonal regulation are necessary for the formation of the arbuscules.

Background: The genus Allium is known for its high chromosomal variability, but most chromosome counts are based on a few individuals and genome size (GS) reports are limited in certain taxonomic groups. This is evident in the Allium sect. Codonoprasum, a species-rich (> 150 species) and taxonomically complex section with weak morphological differences between taxa, the presence of polyploidy and frequent misidentification of taxa.

Introduction: Approximately one-third of all AML patients have a mutation in the Fms-like tyrosine kinase 3 () gene, which is associated with a poor prognosis in these individuals. The 2017 approval of midostaurin, the first FLT3 inhibitor, spurred extensive development of more potent and selective inhibitors with an improved safety profile.

Areas Covered: This review analyzes patent inventions for the treatment of AML using FLT3 inhibitors, covering developments from the earliest to the most recent, disclosed in 2024.

Background: Dexamethasone 6 mg in patients with severe COVID-19 has been shown to decrease mortality and morbidity. The effects of higher doses of corticosteroid, that would further increase anti-inflammatory effects, are uncertain. The objective of our study was to assess the effect of 20 mg dexamethasone vs.

Article Synopsis
  • The study investigates how reallocating time among physical activity, sedentary behavior, and sleep affects obesity indicators like BMI and waist circumference across various age groups.
  • Researchers analyzed data from 9,818 participants using isotemporal substitution models to understand the implications of these behavior changes.
  • Results indicated that even small shifts of 10-30 minutes can significantly impact obesity, with reallocating moderate-to-vigorous physical activity (MVPA) to lighter activities or sedentary behavior having particularly detrimental effects.

The development of disease-modifying therapies (DMTs) for the treatment of multiple sclerosis (MS) has been highly successful in recent decades. It is now widely accepted that early initiation of DMTs after disease onset is associated with a better long-term prognosis. However, the question of when and how to de-escalate or discontinue DMTs remains open and critical.

Sulfonation of IAA in Urtica eliminates its DR5 auxin activity.

Plant Cell Rep

December 2024

Department of Experimental Biology, Palacký University Olomouc, Šlechtitelů 27, CZ-77900, Olomouc, Czech Republic.

N-Sulfonated IAA was discovered as a novel auxin metabolite in Urtica where it is biosynthesized de novo utilizing inorganic sulfate. It showed no auxin activity in DR5::GUS assay, implying possible inactivation/storage mechanism. A novel auxin derivative, N-sulfoindole-3-acetic acid (IAA-N-SOH, SIAA), was discovered in stinging nettle (Urtica dioica) among 116 sulfonated metabolites putatively identified by a semi-targeted UHPLC-QqTOF-MS analysis of 23 plant/algae/fungi species.

Burnout syndrome among general practitioners in the Czech Republic: a repeated survey study.

BMC Prim Care

December 2024

Society of General Practice, J. E. Purkyně Czech Medical Association, Sokolská 490/31, Prague, 12000, Czech Republic.

Background: Given the critical role of general practitioners (GPs), their insufficient medical density and the adverse effects of burnout on both practitioners and the quality of care provided, the prevalence estimates of GP burnout reported in the literature are highly concerning. This nationwide study builds on a 2023 survey that revealed a significant burden of burnout among Czech GPs. The primary objectives were to analyse the prevalence and determinants of burnout and to examine potential trends over time.

We report a study of internal covalent cross-linking with photolytically generated diarylnitrile imines of N-terminal arginine, lysine, and histidine residues in peptide conjugates. Conjugates in which a 4-(2-phenyltetrazol-5-yl)benzoyl group was attached to C-terminal lysine, that we call RAAA--K, KAAA--K, and HAAA--K, were ionized by electrospray and subjected to UV photodissociation (UVPD) at 213 nm. UVPD triggered loss of N and proceeded by covalent cross-linking to nitrile imine intermediates that involved the side chains of N-terminal arginine, lysine, and histidine, as well as the peptide amide groups.

Managing supervisory drift in cognitive behavioral therapy: A narrative review with case vignettes.

Neuro Endocrinol Lett

November 2024

Department of Psychological Sciences, Faculty of Social Science and Health Care, Constantine the Philosopher University, Nitra, Slovak Republic.

Drift is a phenomenon that can occur in cognitive-behavioral supervision, where core components of supervision are omitted, avoided, or deprioritized. This narrative review explores the signs, reasons, and impact of supervisory drift at the experiential, cognitive, and emotional levels for both the supervisor and the supervisee. Additionally, the article presents potential solutions for preventing and addressing supervisory drift, such as staying on track, anticipating problems before they arise, adapting supervision to the supervisee's needs, using active supervision methods to understand drift better, engaging in Supervision of Supervision (SoS), and using alliance measures.

Objectives: This study examined (non-)monotonic time trends in psychological and somatic complaints among adolescents, along with gender differences.

Methods: Repeated cross-sectional Health Behaviour in School-aged Children (HBSC) data from 1994 to 2022 covering 15-year-old adolescents from 41 countries (N = 470,797) were analysed. Three polynomial logistic regression models (linear, quadratic, cubic) were tested for best fit, including separate analyses by gender and health complaints dimension.

Spaced Hybrid TiO/Au Nanotube Arrays with Tailored Optical Properties for Surface-Enhanced Raman Scattering.

ACS Omega

December 2024

Czech Advanced Technology and Research Institute (CATRIN), Regional Centre of Advanced Technologies and Materials Department, Palacký University Olomouc, Šlechtitelů 27, Olomouc 78371, Czech Republic.

Controlling the overall geometry of plasmonic materials allows for tailoring their optical response and the effects that can be exploited to enhance the performance of a wide range of devices. This study demonstrates a simple method to control the size and distribution of gold (Au) nanoparticles grown on the surface of spaced titanium dioxide (TiO) nanotubes by varying the deposition time of magnetron sputtering. While shorter depositions led to small and well-separated Au nanoparticles, longer depositions promoted the formation of quasi-continuous layers with small interparticle gaps.

Introduction And Objectives: With increases in obesity and metabolic syndrome because of lifestyle-related factors, the prevalence of non-alcoholic fatty liver disease (NAFLD) also is increasing worldwide. In a subset of patients with NAFLD, an inflammatory process arises in the steatotic liver, known as non-alcoholic steatohepatitis, that leads to liver fibrosis and liver cirrhosis. In selected patients with obesity, bariatric surgery, and bariatric endoscopy are important therapeutic options.
