Super enhancers are a unique class of enhancers that possess a distinct structure and mechanism, which enable them to exhibit stronger gene transcription regulatory function than classical enhancers, thereby regulating cellular activities. In tumor samples, super enhancers have been identified as crucial players in the development and progression of tumor cells, opening up new avenues for cancer research and treatment. This review provides a concise overview of various models regarding super enhancer assembly and activation, examining the mechanisms through which tumor cells acquire or activate these enhancers and regulate carcinogenic transcription programs.

Background: We developed a prognostic model to evaluate the overall survival (OS) and progression-free survival (PFS) of patients with unresectable hepatocellular carcinoma (u-HCC) treated with Hepatic arterial infusion chemotherapy of infusion oxaliplatin, fluorouracil and leucovorin (FOLFOX-HAIC).
Methods: This model was based on a retrospective study of u-HCC patients treated with the FOLFOX-HAIC (oxaliplatin 130 mg/m, leucovorin 400 mg/m, fluorouracil bolus 400 mg/m on day 1, and fluorouracil infusion 2,400 mg/m for 23-46 h, once every 3-4 weeks). We divided the patients into a training cohort and a validation cohort, used LASSO regression construct prognostic models, predict patient's OS and PFS based on nomograms of models.

Targeted protein delivery with nanocarriers holds significant potential to enhance therapeutic outcomes by precisely directing proteins to specific organs or tissues. However, the complex interactions between nanocarriers and the biological environment pose considerable challenges in designing effective targeted delivery vehicles. In this study, we address this challenge by leveraging DNA-barcoded high-throughput screening.

Reproductive hormones associated with the hypothalamic-pituitary-gonadal (HPG) axis are closely linked to bone homeostasis. In this study, we demonstrate that Gonadotropin inhibitory hormone (GnIH, one of the key reproductive hormones upstream of the HPG axis) plays an indispensable role in regulating bone homeostasis and maintaining bone mass. We find that deficiency of GnIH or its receptor Gpr147 leads to a significant reduction in bone mineral density (BMD) in mice primarily by enhancement of osteoclast activation in vivo and in vitro.

Lactate, a glycolytic intermediate, has a crucial role in cancer metabolism and microenvironment remodeling. Recently, researchers found that lactate mediates lysine lactylation, a novel protein post-translational modification (PTM). Here, we summarize the mechanism and role of lysine lactylation in cancer, and discuss the potential of targeting lysine lactylation in cancer therapy.

A bead-chain metal-organic framework composite was designed and synthesized by assembling a zeolitic imidazolate framework (ZIF) onto manganese dioxide (MnO) nanowires. The prepared catalyst MnO@ZIF-X (X = 1, 2 and 3) was used to facilitate gatifloxacin (GAT) degradation by using potassium peroxymonopulfate (PMS) as an activator. MnO@ZIF-2 exhibited excellent catalytic performance, achieving 100 % degradation of GAT (10 mg/L) in the presence of PMS (1 mM) in 15 min, and the toxicity of the majority of degradation intermediates decreased.

The decline in mitochondrial function has been identified as one of the central pathological mechanisms underlying a variety of aging-related diseases. Nanozymes are nanomaterials with intrinsic enzyme-like properties and are important alternatives to natural enzymes. As emerging biocatalysts, nanozymes exhibit significant potential in mimicking the activity of natural enzymes, enhancing mitochondrial function, and offering novel therapeutic strategies for aging-related conditions.
