The gabapentinoids, gabapentin, and pregabalin, target the αδ subunits of voltage-gated calcium channels. Initially licensed for pain and seizures, they have become widely prescribed drugs. Many of these uses are off-label for psychiatric indications, and there is increasing concern about their safety, so it is particularly important to have good evidence to justify this usage.

Article Synopsis
  • Training the innate immune system with oral bacterial extracts like Broncho-Vaxom (BV) OM-85 shows positive effects on infection recovery and lung health.
  • In studies using mice infected with a coronavirus, BV OM-85 treatment resulted in more macrophages in the lungs, leading to an accelerated immune response and better protection against infection.
  • The findings also indicate that BV OM-85 helps the lungs return to a healthy state more quickly after infection, and this effect relies on the activation of type I interferon signaling.

Purpose: Both brodalumab and guselkumab improve skin clearance in patients with moderate-to-severe plaque psoriasis after inadequate response to ustekinumab. In the absence of a direct head-to-head comparison, the relative efficacy of brodalumab and guselkumab in non-responders to ustekinumab were compared using a matching-adjusted indirect comparison (MAIC).

Patients And Methods: Individual patient data for brodalumab (n = 121) were pooled from the AMAGINE-2 and -3 trials and adjusted using a propensity score reweighting method, so that baseline and week 16 characteristics matched the aggregate published data of patients with an inadequate response to ustekinumab who switched to guselkumab (n = 135) in the NAVIGATE trial.

Background: EUS-guided fine-needle aspiration (EUS-FNA) has emerged as the primary modality for the cytologic diagnosis of pancreatic solid masses. The aim of this study is to determine whether technical factors including suction (S), non-suction (NS), capillary sampling with stylet slow-pull (CSSS), and the number of needle actuations (to-and-fro needle movements) may affect the accuracy of EUS-FNA for pancreatic solid masses at facilities without on-site cytopathology.

Methods: The diagnostic yield of malignancy, blood contamination and cellularity at each sample acquired from EUS-FNA with or without S and different numbers of actuation (10, 15 and 20) were measured (study I).

Medical imaging is a central part of clinical diagnosis and treatment guidance. Machine learning has increasingly gained relevance because it captures features of disease and treatment response that are relevant for therapeutic decision-making. In clinical practice, the continuous progress of image acquisition technology or diagnostic procedures, the diversity of scanners, and evolving imaging protocols hamper the utility of machine learning, as prediction accuracy on new data deteriorates, or models become outdated due to these domain shifts.

Bioanalytical Sciences, BioMarin Pharmaceutical, 791 Lincoln Avenue, San Rafael, CA 94901, USA.

Adeno-associated virus (AAV)-based gene therapies have recently shown promise as a novel treatment for hereditary diseases. Due to the viral origin of the vector capsid, however, cellular immune response may be elicited that could eliminate transduced target cells. To monitor cellular immune responses in clinical trials, we optimized and bioanalytically validated a sensitive, robust, and reliable interferon-γ (IFN-γ) enzyme-linked immunospot (ELISpot) assay.
