Amyloid-β aggregates activate peripheral monocytes in mild cognitive impairment.

Nat Commun

February 2024

Department of Biomedicine, Aarhus University, The Skou Building, Høegh-Guldbergs Gade 10, DK-8000, Aarhus C, Denmark.

Article Synopsis
  • The peripheral immune system influences neurodegenerative diseases like Alzheimer's by both protecting the brain and causing inflammation; however, the exact mechanisms are not fully understood.
  • Researchers found large Aβ aggregates in the plasma of patients with mild cognitive impairment, which are linked to early signs of Alzheimer's pathology and a decrease in certain monocytes.
  • The study identifies complement receptor 4 as a key player in interacting with amyloids, enhancing phagocytosis and lysosomal activity in microglia, and suggests that Aβ aggregates may help recruit immune cells into the brain, affecting the disease process.

The discovery of a copper precatalyst that facilitates the key mechanistic steps of arene halodeboronation has allowed a step change in the synthesis of radioiodine-containing arenes. The active precatalyst [Cu(OAc)(phen)]OAc was shown to perform room temperature radio-iododeboronation of aryl boronic acids with 1-2 mol % loadings and 10 min reaction times. These mild conditions enable particularly clean reactions, as demonstrated with the efficient preparation of the radiopharmaceutical and SPECT tracer, -iodobenzylguanidine (MIBG).

Background And Purpose: Carotid artery stenosis or occlusion (CASO) is a causative disease of vascular cognitive impairment (VCI) attributed to cerebral hypoperfusion, even without the development of symptomatic ischemic stroke. Preclinically, resveratrol has been demonstrated to play an important role in improving cognitive function in rodent CASO models. This study investigated the association between long-term resveratrol intake and improvements in cognitive and cerebral hemodynamic impairments in patients with CASO.

Article Synopsis
  • The study examines how early treatment responses impact the outcomes for patients with locally advanced non-small cell lung cancer (LA-NSCLC) receiving concurrent chemo-radiotherapy (cCRT), focusing specifically on the effects of induction chemotherapy (iCX) on patterns of failure (PoF).
  • Researchers analyzed data from 276 patients to identify correlations between treatment response metrics—such as tumor volume and SUVpeak—and the likelihood of loco-regional or distant failures.
  • Results indicated that squamous cell carcinoma (SCC) patients were more likely to experience loco-regional failures, whereas non-SCC patients had a higher risk of distant failures; also, higher SUVpeak values and poor tumor volume response were linked to increased failure risk.

Deposition of amyloid beta (Aβ) into plaques is a major hallmark of Alzheimer's disease (AD). Different amyloid precursor protein (APP) mutations cause early-onset AD by altering the production or aggregation properties of Aβ. We recently identified the Uppsala APP mutation (APPUpp), which causes Aβ pathology by a triple mechanism: increased β-secretase and altered α-secretase APP cleavage, leading to increased formation of a unique Aβ conformer that rapidly aggregates and deposits in the brain.

The 18 kDa translocator protein is a well-known biomarker of neuroinflammation, but also plays a role in homeostasis. PET with 18 kDa translocator protein radiotracers [C]PBR28 in humans and [F]GE180 in mice has demonstrated sex-dependent uptake patterns in the healthy brain, suggesting sex-dependent 18 kDa translocator protein expression, although humans and mice had differing results. This study aimed to assess whether the 18 kDa translocator protein PET radiotracer [F]LW223 exhibited sexually dimorphic uptake in healthy murine brain and peripheral organs.

Emotions, bodily sensations and movement are integral parts of musical experiences. Yet, it remains unknown i) whether emotional connotations and structural features of music elicit discrete bodily sensations and ii) whether these sensations are culturally consistent. We addressed these questions in a cross-cultural study with Western (European and North American, n = 903) and East Asian (Chinese, n = 1035).

Evidence on the long-term effects of reducing sedentary behaviour (SB) on blood pressure (BP) is scarce. Therefore, we performed a sub-analysis of the BP effects of a six-month intervention that aimed at reducing SB by 1 h/day and replacing it with non-exercise activities. Sixty-four physically inactive and sedentary adults with metabolic syndrome (58% female, 58 [SD 7] years, BP 143/88 [16/9] mmHg, SB 10 [1] h/day) were randomised into intervention (INT, n = 33) and control (CON, n = 31) groups.

CSF p-tau205: a biomarker of tau pathology in Alzheimer's disease.

Acta Neuropathol

January 2024

Department of Psychiatry and Neurochemistry, Institute of Neuroscience and Physiology, The Sahlgrenska Academy at the University of Gothenburg, Mölndal, Sweden.

Post-mortem staging of Alzheimer's disease (AD) neurofibrillary pathology is commonly performed by immunohistochemistry using AT8 antibody for phosphorylated tau (p-tau) at positions 202/205. Thus, quantification of p-tau205 and p-tau202 in cerebrospinal fluid (CSF) should be more reflective of neurofibrillary tangles in AD than other p-tau epitopes. We developed two novel Simoa immunoassays for CSF p-tau205 and p-tau202 and measured these phosphorylations in three independent cohorts encompassing the AD continuum, non-AD cases and cognitively unimpaired participants: a discovery cohort (n = 47), an unselected clinical cohort (n = 212) and a research cohort well-characterized by fluid and imaging biomarkers (n = 262).

Purpose: Simultaneous PET-MRI improves inflammatory cardiac disease diagnosis. However, challenges persist in respiratory motion and mis-registration between free-breathing 3D PET and 2D breath-held MR images. We propose a free-breathing non-rigid motion-compensated 3D T -mapping sequence enabling whole-heart myocardial tissue characterization in a hybrid 3T PET-MR system and provides non-rigid respiratory motion fields to correct also simultaneously acquired PET data.
