Suppression of Dad1 induces cardiomyocyte death by weakening cell adhesion.

Am J Physiol Cell Physiol

January 2025

Laboratory of Clinical Science and Biomedicine, Graduate School of Pharmaceutical Sciences, Osaka University, Suita City, Japan.

As cardiomyocyte loss causes heart failure, inhibition of cardiomyocyte death may be a therapeutic strategy against heart failure. In this study, we have identified defender against cell death 1 (Dad1) as a candidate regulator of cardiomyocyte death, using complementary DNA microarray and siRNA knockdown screening. Dad1 is a subunit of oligosaccharyltransferase (OST) complex that is responsible for protein N-glycosylation; however, its function in cardiomyocytes remains unknown.

Department of Pharmacy, Osaka University Hospital, Suita, Osaka 5650871, Japan; Department of Hospital Pharmacy, Graduate School of Medicine, Osaka University, Suita, Osaka 5650871, Japan.

Article Synopsis
  • - Dasatinib is a medication used to treat chronic myeloid leukemia (CML), but some patients develop resistance due to high levels of the ABCG2 protein in their cancer cells.
  • - A study involving 65 CML patients found that those taking the xanthine oxidase inhibitor febuxostat alongside dasatinib had better treatment outcomes, with significantly higher rates of early and major molecular response.
  • - Laboratory experiments showed that febuxostat can lower cell viability and increase dasatinib levels in CML cells, suggesting it helps enhance dasatinib's effects by reducing ABCG2's ability to eject the drug from cells, which may offer new ways to tackle TKI resistance in CML treatment. *

Article Synopsis
  • Pareidolias are perceptual deficits where people see meaningful shapes in ambiguous visuals, studied in neurodegenerative diseases using a noise pareidolia test.
  • This research explored differences between paper-based and digital testing methods for pareidolias among healthy individuals and patients with Alzheimer's, DLB, and Parkinson's disease.
  • Results show that pareidolic perceptions remain consistent in both testing formats, and digital testing on smartphones can effectively maintain the functionality of assessing these visual illusions.

Background Advances in imaging technology and the increased use of abdominal imaging have led to a rise in renal cell carcinoma (RCC) detection. While surgery remains the primary treatment for small RCCs, minimally invasive procedures like cryoablation are gaining popularity, particularly for patients with comorbidities or renal dysfunction. CT-guided cryoablation offers advantages, including high spatial resolution and real-time visualization during the procedure.
