Arachidonic acid regulates pluripotency by modulating cellular energetics via fatty acid synthesis and mitochondrial fission.

Biochem Biophys Res Commun

December 2024

Stem Cells and Regenerative Medicine Centre, Yenepoya Research Centre, Yenepoya (Deemed to be University), Mangalore 575018, India. Electronic address:

Article Synopsis
  • Arachidonic acid (AA) is a key omega-6 fatty acid that influences various cellular functions and is crucial for the efficiency of converting somatic cells into induced pluripotent stem cells (iPSCs).
  • Research shows that lower concentrations of AA enhance pluripotency in human induced pluripotent stem cells (hiPSCs) by increasing the expression of pluripotency markers and promoting fatty acid synthesis and mitochondrial fission.
  • However, higher concentrations of AA lead to differentiation of stem cells, indicating a concentration-dependent effect on stem cell behavior, with potential regulatory mechanisms involving interactions with enzymes ACC1 and CREB1.

The Dysregulation of Essential Fatty Acid (EFA) Metabolism May Be a Factor in the Pathogenesis of Sepsis.

Medicina (Kaunas)

June 2024

UND Life Sciences, 2221 NW 5th St., Battle Ground, WA 98604, USA.

Article Synopsis
  • A deficiency in essential fatty acids (EFAs) and changes in their metabolism may significantly contribute to the development and severity of sepsis, influencing mortality rates.
  • The ineffectiveness of treatments like corticosteroids and certain monoclonal antibodies in sepsis can be linked to these altered EFA metabolic pathways.
  • Restoring normal EFA metabolism could lead to new therapeutic strategies for preventing and managing sepsis and other serious conditions by balancing inflammatory responses.

PROTACs in the Management of Prostate Cancer.

Molecules

April 2023

Department of Chemiinformatics, Centella Scientific, JHUB, Jawaharlal Nehru Technological University, Hyderabad 500085, India.

Cancer treatments with targeted therapy have gained immense interest due to their low levels of toxicity and high selectivity. Proteolysis-Targeting Chimeras (PROTACs) have drawn special attention in the development of cancer therapeutics owing to their unique mechanism of action, their ability to target undruggable proteins, and their focused target engagement. PROTACs selectively degrade the target protein through the ubiquitin-proteasome system, which describes a different mode of action compared to conventional small-molecule inhibitors or even antibodies.

Background: Alterations in the levels of nutrients like calcium, ferritin, and electrolytes play a pivotal role in human physiology and might serve as biomarkers. Ferritin, an iron storage protein is important in various metabolic reactions of both cancer and cancer stem cells (CSCs) and is found to regulate 'stemness' leading to cancer relapse. Interestingly, ferritin levels are found to be regulated by calcium uptake.
