Follicular dendritic cells (FDCs) play a crucial role in generating high-affinity antibody-producing B cells during the germinal center (GC) reaction. Herein, we analysed the altered gene expression profile of a mouse FDC line, FL-Y, following lymphotoxin β receptor stimulation, and observed increased Slam-family member 8 (Slamf8) mRNA expression. Forced Slamf8 expression and SLAMF8-Fc addition enhanced the ability of FL-Y cells to induce FDC-induced monocytic cell (FDMC) differentiation.

Nationwide in Japan, a community-based vision-screening program in 3.5-year-old children is conducted in three steps: questionnaires and home visual acuity testing as the primary screening; visual acuity testing by nurses and pediatricians' inspection in community health centers as the secondary screening; and examinations by ophthalmologists as the tertiary screening. In this study, we introduced photorefraction with a Spot vision screener in addition to visual acuity testing to answer the clinical question of whether photorefraction could better detect eye diseases and potentially replace visual acuity testing.

Cognitive Neuroscience Laboratory, Graduate School of Interdisciplinary Science and Engineering in Health Systems, Okayama University, Okayama, Japan.

Evidence has shown that multisensory integration benefits to unisensory perception performance are asymmetric and that auditory perception performance can receive more multisensory benefits, especially when the attention focus is directed toward a task-irrelevant visual stimulus. At present, whether the benefits of semantically (in)congruent multisensory integration with modal-based attention for subsequent unisensory short-term memory (STM) retrieval are also asymmetric remains unclear. Using a delayed matching-to-sample paradigm, the present study investigated this issue by manipulating the attention focus during multisensory memory encoding.

Design and Synthesis of Glycosylated Cholera Toxin B Subunit as a Tracer of Glycoprotein Trafficking in Organelles of Living Cells.

Chemistry

July 2022

Graduate School of Interdisciplinary Science and Engineering in Health Systems, Okayama University, 3-1-1 Tsushimanaka, Okayama, 700-8530, Japan.

Glycosylation of proteins is known to be essential for changing biological activity and stability of glycoproteins on the cell surfaces and in body fluids. Delivering of homogeneous glycoproteins into the endoplasmic reticulum (ER) and the Golgi apparatus would enable us to investigate the function of asparagine-linked (N-) glycans in the organelles. In this work, we designed and synthesized an intentionally glycosylated cholera toxin B-subunit (CTB) to be transported to the organelles of mammalian cells.

Emerging evidence suggests white matter network abnormalities in patients with schizophrenia (SZ) and bipolar disorder (BD), but the alterations in dynamics of the white matter network in patients with SZ and BD are largely unknown. The white matter network of patients with SZ (n = 45) and BD (n = 47) and that of healthy controls (HC, n = 105) were constructed. We used dynamics network control theory to quantify the dynamics metrics of the network, including controllability and synchronizability, to measure the ability to transfer between different states.

Calcium/calmodulin-dependent protein kinase kinases (CaMKKs) activate CaMKI, CaMKIV, protein kinase B/Akt, and AMP-activated protein kinase (AMPK) by phosphorylating Thr residues in activation loops to mediate various Ca -signaling pathways. Mammalian cells expressing CaMKKα and CaMKKβ lacking Arg/Pro-rich insert domain (RP-domain) sequences showed impaired phosphorylation of AMPKα, CaMKIα, and CaMKIV, whereas the autophosphorylation activities of CaMKK mutants remained intact and were similar to those of wild-type CaMKKs. Liver kinase B1 (LKB1, an AMPK kinase) complexed with STRAD and MO25 and was unable to phosphorylate CaMKIα and CaMKIV; however, mutant LKB1 with the RP-domain sequences of CaMKKα and CaMKKβ inserted between kinase subdomains II and III acquired CaMKIα and CaMKIV phosphorylating activity in vitro and in transfected cultured cells.

Tactile angle discriminability improvement: contributions of working memory training and continuous attended sensory input.

J Neurophysiol

Graduate School of Interdisciplinary Science and Engineering in Health Systems, Okayama University, Okayama, Japan.

Perceptual learning is commonly assumed to enhance perception through continuous attended sensory input. However, learning is generalizable to performance in untrained stimuli and tasks. Although previous studies have observed a possible generalization effect across tasks as a result of working memory (WM) training, comparisons of the contributions of WM training and continuous attended sensory input to perceptual learning generalization are still rare.

Department of Psychology, Research Center for Psychology and Behavioral Sciences, Soochow University, Suzhou, China; Laboratory, Graduate School of Interdisciplinary Science and Engineering In Health Systems, Okayama University, Okayama, Japan.

Pairing a single visual stimulus with multiple auditory stimuli will lead to the illusory perception of multiple visual stimuli, which is known as sound-induced flash illusion (SIFI). The present study adopted the classic SIFI paradigm to investigate whether value-associated tasks could affect the SIFI. By adjusting the sequence of reward and nonreward conditions, we also examined the effect of reward history on SIFI.

Neuropsychological studies have demonstrated that the preferential processing of near-space and egocentric representation is associated with the self-prioritization effect (SPE). However, relatively little is known concerning whether the SPE is superior to the representation of egocentric frames or near-space processing in the interaction between spatial reference frames and spatial domains. The present study adopted the variant of the shape-label matching task (i.

Division of Molecular and Metabolic Medicine, Graduate School of Medicine, Kobe University, 7-5-1 Kusunoki-cho, Chuo-ku, Kobe, Hyogo, 650-0017, Japan.

Patients with type 2 diabetes often exhibit impairments in both glucose-induced insulin secretion (GIIS) and incretin-induced insulin secretion (IIIS). These phenotypes are associated with altered glucose metabolism in pancreatic β-cells, although the molecular mechanisms remain unclear. Here, we used MIN6-K8 pancreatic β-cell lines as a model to examine the effect of O-linked N-acetylglucosamine glycosylation (O-GlcNAcylation), a glucose-induced protein posttranslational modification, on insulin secretion.

Cognitive Neuroscience Laboratory, Graduate School of Interdisciplinary Science and Engineering in Health Systems, Okayama University, Okayama, 7008530, Japan.

Neuronal studies have shown that selectively attending to a common object in one sensory modality results in facilitated processing of that object's representations in the ignored sensory modality. Thus, the audiovisual (AV) integration of common objects can be observed under modality-specific selective attention. However, little is known about whether this AV integration can also occur under increased attentional load conditions.

Cancer-inducing niche: the force of chronic inflammation.

Br J Cancer

July 2022

Department of Biotechnology and Drug Discovery, Graduate School of Interdisciplinary Science and Engineering in Health Systems, Okayama University, Okayama, 700-8530, Japan.

The growth of cancer tissue is thought to be considered driven by a small subpopulation of cells, so-called cancer stem cells (CSCs). CSCs are located at the apex of a hierarchy in a cancer tissue with self-renewal, differentiation and tumorigenic potential that produce the progeny in the tissue. Although CSCs are generally believed to play a critical role in the growth, metastasis, and recurrence of cancers, the origin of CSCs remains to be reconsidered.

Diphenyleneiodonium efficiently inhibits the characteristics of a cancer stem cell model derived from induced pluripotent stem cells.

Cell Biochem Funct

April 2022

Department of Biotechnology and Drug Discovery, Graduate School of Interdisciplinary Science and Engineering in Health Systems, Okayama University, Okayama, Japan.

Article Synopsis
  • Diphenyleneiodonium (DPI) has been reevaluated as a potential anticancer drug, specifically targeting cancer stem cells (CSCs) which are often resistant to standard chemotherapy.
  • The study found that DPI is effective at much lower concentrations (around 5.52 to 12 nM) in three-dimensional culture assays compared to traditional assays, where higher concentrations (712 nM) were required for noticeable effects.
  • The research suggests that 3D culture systems may provide a better model for screening anti-CSC drug candidates, as they more accurately replicate the tumor microenvironment compared to 2D culture systems.

Article Synopsis
  • - The study identifies two sperm-specific polypeptides, Kastor and Polluks, produced by a mouse gene initially thought to only encode a long noncoding RNA.
  • - Both Kastor and Polluks interact with the voltage-dependent anion channel (VDAC) located in the outer mitochondrial membrane, which is crucial for sperm motility.
  • - Deficits in either Kastor or Polluks severely impact male fertility by resulting in abnormal mitochondrial structures in sperm, suggesting that their combined function is vital for proper sperm development and function.

Aldehyde dehydrogenases (ALDHs) are the major enzyme superfamily for the aldehyde metabolism. Since the ALDH polymorphism leads to the accumulation of acetaldehyde, we considered that the enhancement of the liver ALDH activity by certain food ingredients could help prevent alcohol-induced chronic diseases. Here, we evaluated the modulating effects of 3-hydroxyphenylacetic acid (OPAC), the major metabolite of quercetin glycosides, on the ALDH activity and acetaldehyde-induced cytotoxicity in the cultured cell models.

The frontoparietal network (FPN), including bilateral frontal eye field, inferior parietal sulcus, and supplementary motor area, has been linked to attention processing, including spatiotemporal and sensory modality domains. However, it is unclear whether FPN encodes representations of these domains that are generalizable across subdomains. We decomposed multivariate patterns of functional magnetic resonance imaging activity from 20 participants into domain-specific components and identified latent multivariate representations that generalized across subdomains.
