Development of different platforms would be useful for designing functional antibodies to improve the efficiency of antibody-based drugs. Three-dimensional domain swapping (3D-DS) may occur in the variable region of antibody light chain #4C214A, and a pair of domain-swapped dimers may interact with each other to form a tetramer. In this study, to stabilize the 3D-DS dimer structure in #4C214A, Val2 in strand A (swapping region) and Thr97 in strand G were replaced with Cys residues, generating #4 V2C/T97C/C214A with a Cys2-Cys97 disulfide bond that cross-links strands A and G of different protomers.

Unmethylated cytosine-phosphate-guanine (CpG) motifs are often found in bacteria and viruses, but are rare in mammals. In mammals, CpG oligodeoxynucleotides (CpG ODN) stimulate the innate immune system via toll-like receptor 9 (TLR9). However, TLR9 is absent in birds; instead, TLR21 serves as the receptor for CpG ODN.

Purpose: Brain iron deficiency is known to cause dopaminergic dysfunction in people with restless legs syndrome. Considering the role that dopamine plays in the wake-promoting system, we speculated that iron deficiency may contribute to the development of excessive daytime sleepiness (EDS). Therefore, we conducted the present study to examine the association between iron intake and EDS in the general population.

Landscapes are shaped by tectonic, climatic, and surface processes over geological timescales, but we rarely witness the events of marked landscape change. The moment magnitude 7.5 Noto Peninsula earthquake in central Japan was caused by a large thrust faulting, up to nearly 10 meters of slip, that expanded more than 150 kilometers along the fault zone.

Article Synopsis
  • The study examines how bariatric/metabolic surgery affects the incidence and severity of COVID-19 among patients in Japan, amidst ongoing debates about its protective benefits.
  • Data were collected from 83 institutions, revealing that while there were no severe cases or deaths among 119 patients studied, moderate COVID-19 cases were notably more prevalent in these bariatric patients than in the general population.
  • The findings suggest that bariatric/metabolic surgery does not reduce the risk of COVID-19 complications, as moderate cases were linked to specific risk factors such as dyslipidemia and prior infection.

Organic anion transporting polypeptides (OATP) 1B1 and 1B3 are expressed in liver cells and are involved in drug uptake in the liver. OATP1B activity varies due to polymorphisms and is decreased by OATP1B inhibitors. Variability of OATP1B activity impacts the pharmacokinetics of OATP1B substrate drugs through drug-drug interactions.

Department of Microbiology, Faculty of Medicine and Research Center for Global and Local Infectious Diseases, Oita University, Oita, Japan.

Article Synopsis
  • Rabies is a highly infectious viral disease that mainly affects the central nervous system, is often transmitted by mammals like dogs, has a long incubation period, and carries a nearly 100% mortality rate with no effective treatment available.
  • Japan eradicated rabies in 1957 through stringent vaccination measures, and the country maintains strict controls that surpass international standards, despite experiencing a few imported cases since then.
  • Recent events, such as the quarantine of evacuee dogs from Ukraine and a dog bite incident in Gunma Prefecture that raised concerns about rabies, highlight ongoing vigilance and the need for effective rabies control and management strategies both in Japan and globally.
