This study was aimed to explore the effects of STI571 alone or with As₂O₃ on proliferation, apoptosis and caspase 3, bcl-xL mRNA expression of K562 cells, and the molecular mechanism of As₂O₃ enhancing the anti-leukemia effect of STI571 so as to provide the scientific basis for clinical treatment of chronic myeloid leukemia. The effect of drugs on proliferation of K562 cells was assayed by MTT method, the apoptosis rate of K562 cells was detected by flow cytometry with Annexin V/PI double staining, the caspase 3, bcl-xL mRNA expressions of K562 cells were determined by real time quantitative PCR. The results showed that STI571 alone or with As₂O₃ both could inhibit the proliferation of K562 cells.

Front Biosci (Elite Ed)

June 2010

Department of Life Science, North China Coal medical college, No 57 JianShe South Road, Tangshan 063000, China.

The transcription factor, AP-1, plays an important role in cellular proliferation, transformation and death. We previously showed that AC3-33 (GenBank name: c3orf33, FLJ31139), significantly inhibited transcriptional activity of AP-1. In this study, we report a method to express and purify AC3-33 in E.

Article Synopsis
  • The study investigates the protective effects of edaravone on severe traumatic brain injury (TBI) in rats, aiming to understand its underlying mechanisms.
  • Two hundred and seventy-three male rats were divided into four groups: control, model (TBI), low-dose, and high-dose edaravone treatment, with various assessments made on brain tissue damage and neuron apoptosis over time.
  • Results indicated that edaravone treatment reduced neuronal damage, apoptosis, and improved learning and memory function compared to untreated TBI rats, suggesting a therapeutic potential for edaravone in TBI recovery.
