The aim of this study was to evaluate the effect of different concentrations of triciribine, a selective Akt inhibitor, on various aspects of oocyte maturation and on the IVF of bovine embryos. Cumulus-oocyte complexes (COCs) were matured in vitro in medium supplemented with: 0 (control), 1, 5, 10, and 20 μM of triciribine. The nuclear maturation was assessed by staining with acetic orcein, while the cytoplasmic maturation was evaluated by mitochondrial (MitoTracker® Red CMXRos) and lipid droplets distribution (LipidTOX).

Ten male sheep (Sheep group; SGk) and seven male goats (Goat group; GGks+) were used in this study. The objective was to compare the use of racemic ketamine or ketamine S(+) associated with lidocaine on spinal anesthesia and evaluate if the drugs leads to a surgical anesthesia state, as well as to verify the cardiorespiratory, sedative and motor effects of this technique in these species. After correct placement of the needle in the subarachnoid space, 3.

Rapid Characterization of Triterpene Saponins from by HPLC-ESI-MS/MS.

Molecules

July 2019

Graduate Program in Natural and Synthetic Bioactive Products, Health Sciences Center, Universidade Federal da Paraíba, João Pessoa 58051-900, Paraíba, Brazil.

Vogel (Leguminosae) is a species popularly known in Brazil as "urinária", "urinana", and "carrapicho", it is popularly used as a diuretic and in the treatment of venereal diseases. A specific methodology to obtain a saponin-enriched fraction and high-performance liquid chromatography coupled with diode array detection, ion trap mass spectrometry, and TOF-MS (HPLC-DAD-ESI-MS/MS) was applied for the analysis of triterpene saponins. The MS and MS/MS experiments were carried out by ionization in negative mode.

Variants in the CYP19A1 gene can affect in vitro embryo production traits in cattle.

J Assist Reprod Genet

December 2018

Embrapa Dairy Cattle Research Unit, Laboratory for Animal Reproduction, Santa Mônica Experimental Farm (CESM), Valença, Rio de Janeiro, Brazil.

Purpose: This study aimed to associate DNA variants in promoter and exon flanking regions of the CYP19A1 gene with in vitro embryo production traits in cattle. The role of transcription factor binding sites created or lost due to DNA sequence variation and their possible effect on gene expression was also evaluated.

Methods: We collected date from Gyr dairy oocyte donor cows (Bos taurus indicus) at a commercial in vitro embryo production farm and analyzed the genotype-phenotype association with in vitro production traits.

Surface temperature of ewes during estrous cycle measured by infrared thermography.

Theriogenology

October 2018

Laboratory for Animal Breeding and Genetic Improvement, Norte Fluminense State University, Av. Alberto Lamego, 2000, CEP: 28013-602, Campos dos Goytacazes, Rio de Janeiro, Brazil. Electronic address:

The use of infrared thermography has not been used previously to assist sheep reproduction. The aim of this study was evaluate the viability of infrared thermography to identify body surface temperature patterns during ewes' estrous cycle. The estrous cycles of 20 Santa Ines ewes were synchronized through hormone treatment (progesterone implant).
