Division of Gastroenterology and Hepatology, Department of Medicine, University of Illinois at Chicago, Chicago, Illinois, USA.

Sickle cell disease (SCD) is one of the most common genetic disorders in the world predominantly affecting economically disadvantaged populations. There is a notable discrepancy between the growing adult SCD population and available diagnostic and therapeutic interventions for SCD. Sickle cell hepatopathy (SCH) is an all-inclusive term to describe the acute and chronic liver manifestations of SCD.

  • - Atrioventricular block (AVB) is uncommon in hyperthyroidism, and this study aimed to clarify its prevalence, types, management, and outcomes through a systematic review of existing literature.
  • - The review included 56 studies with 87 patients, revealing that complete heart block (CHB) was the most frequent form of AVB, affecting over 51% of those studied, and only a small number required pacemaker insertion.
  • - Findings indicate that while CHB is prevalent, most cases in hyperthyroid patients can be treated effectively with anti-thyroid medications, with limited differences in outcomes based on whether patients received pacing.

Objective: This study aims to utilize PEGylated poly (lactic-co-glycolic acid) (PLGA) nanoparticles as a delivery system for simultaneous administration of the BRAF peptide, a tumor-specific antigen, and imiquimod (IMQ). The objective is to stimulate dendritic cell (DC) maturation, activate macrophages, and facilitate antigen presentation in C57BL6 mice.

Methods: PEG-PLGA-IMQ-BRAF nanoparticles were synthesized using a PLGA-PEG-PLGA tri-block copolymer, BRAF, and IMQ.

Background And Objectives: Acute appendicitis is one of the most commonly encountered surgical emergencies on a global level. Due to the requirement of an immediate clinical diagnosis and the presence of limited resources, clinicians and diagnosticians refer to scoring systems to diagnose this condition, among which Alvarado and Tzanakis scoring systems are widely used. This meta-analysis aims to compare the diagnostic accuracy of these two systems.

Establishing clinically meaningful changes in pain experiences remains important for clinical trials of chronic pain treatments. Regulatory guidance and pain measurement initiatives have recommended including patient-reported global assessment measures (eg, Patient-Global Impression of Change [PGIC]) to aid interpretation of within-patient differences in domain-specific clinical trial outcomes (eg, pain intensity). The objectives of this systematic review were to determine the frequency of global assessment measures inclusion, types of measures, domains assessed, number and types of response options, and how measures were analyzed.

Introduction: Portal vein thrombosis (PVT) is a rare medical condition that obstructs blood flow in the portal vein, with cirrhosis as a common predisposing factor. However, its association with oral contraceptive pills (OCPs), particularly with progestins, remains inadequately explored. This case report aims to contribute to this understanding, focusing on the rare presentation of PVT-induced intestinal obstruction in a female on prolonged OCP therapy.

  • Transgenic Arabidopsis thaliana plants were created by introducing a gene for fructose-1,6-bisphosphatase (FBPase), resulting in enhanced plant growth and stability.
  • The AtFBPase plants showed significant improvements in height (35cm), leaf count (25), chlorophyll levels (28%), water use efficiency (increased from 1.5 to 2.6 μmol CO2 μmol-1 H2O), and stomatal conductance (20%).
  • These enhancements contributed to a higher photosynthetic rate (2.7 μmol m-2 s-1), indicating FBPase's crucial role in boosting photosynthesis, which may benefit economically important crops in

  • Pakistan has experienced multiple HIV outbreaks over the last two decades, particularly centered in Larkana, which has seen alarming cases shift from high-risk groups to a low-risk pediatric population in 2019.
  • This shift raises concerns about the increasing spread of HIV into the general public and suggests that control strategies are not effectively managing the epidemic, leading to a rise in overall cases.
  • The review aims to summarize known risk factors from past outbreaks, investigate potential connections like child sexual abuse in recent cases, and propose strategies to combat future HIV outbreaks in Pakistan.

  • NAFLD is characterized by fat buildup in the liver without other causes, and the study investigates how HbA1c (a marker for blood sugar control) correlates with liver enzyme ALT levels in patients with type 2 diabetes.
  • The study included 130 T2DM patients, finding that 60% had uncontrolled diabetes, with elevated ALT levels in both genders, significantly higher in those with poor glycemic control.
  • Results indicate a positive relationship between high HbA1c and increased ALT levels, suggesting that monitoring these levels can help assess the severity of NAFLD in T2DM patients.

APOE2 gene therapy reduces amyloid deposition and improves markers of neuroinflammation and neurodegeneration in a mouse model of Alzheimer disease.

Mol Ther

May 2024

Alzheimer Research Unit, Massachusetts General Hospital Institute for Neurodegenerative Disease, Charlestown, MA 02129, USA; Department of Neurology, Massachusetts General Hospital and NeuroDiscovery Center, Harvard Medical School, Boston, MA 02114, USA.

Epidemiological studies show that individuals who carry the relatively uncommon APOE ε2 allele rarely develop Alzheimer disease, and if they do, they have a later age of onset, milder clinical course, and less severe neuropathological findings than people without this allele. The contrast is especially stark when compared with the major genetic risk factor for Alzheimer disease, APOE ε4, which has an age of onset several decades earlier, a more aggressive clinical course and more severe neuropathological findings, especially in terms of the amount of amyloid deposition. Here, we demonstrate that brain exposure to APOE ε2 via a gene therapy approach, which bathes the entire cortical mantle in the gene product after transduction of the ependyma, reduces Aβ plaque deposition, neurodegenerative synaptic loss, and, remarkably, reduces microglial activation in an APP/PS1 mouse model despite continued expression of human APOE ε4.
