The escalating prevalence of skeletal muscle disorders highlights the critical need for innovative treatments for severe injuries such as volumetric muscle loss. Traditional treatments, such as autologous transplants, are constrained by limited availability and current scaffolds often fail to meet complex clinical needs. However, the biological effects of allosteric driver mutations during tumorigenesis are mostly unknown. Here, we mapped somatic mutations generated from 10 tumor-normal matched HNSCC samples into allosteric sites to prioritize the mutated allosteric proteins via whole-exome sequencing and AlloDriver, identifying the specific mutation H351Q in β-glucuronidase (GUSB), a lysosomal enzyme, as a novel allosteric driver mutation, which considerably encouraged HNSCC progression both in vitro and in vivo.

All-solid-state lithium metal batteries are regarded as next-generation devices for energy storage due to their safety and high energy density. The issues of lithium dendrites and poor mechanical compatibility with electrodes present the need for developing solid-state electrolytes with high stiffness and damping, but it is a contradictory relationship. Here, inspired by the superstructure of tooth enamel, we develop a composite solid-state electrolyte composed of amorphous ceramic nanotube arrays intertwined with solid polymer electrolytes.

Although existing reports indicating that mixed lineage kinase domain-like protein (MLKL) is involved in macrophage necroptosis and inflammasome activation, the downstream mechanism of MLKL in necroptosis remain poorly characterized in AMI.

Inducing the degradation of KRAS represents a novel strategy to combat cancers with KRAS mutation. In this study, we identify ubiquitin-specific protease 2 (USP2) as a novel deubiquitinating enzyme of KRAS in multiple myeloma (MM). Specifically, we demonstrate that gambogic acid (GA) forms a covalent bond with the cysteine 284 residue of USP2 through an allosteric pocket, inhibiting its deubiquitinating activity.

Aim: To evaluate the diagnostic accuracy of an active matrix metalloproteinase-8 (aMMP-8) point-of-care oral rinse test (POC-ORT) for predicting periodontitis in treatment-naïve subjects in two independent studies and update a recent meta-analysis.
Methods: The aMMP-8 POC-ORT index test was performed in a representative population in Hong Kong, China, and a consecutive convenience sample in Shanghai, China. The reference standard was the 2017 World Workshop classification of periodontal diseases.
