Background: Minimally verbal children with autism are understudied and lack effective treatment options. Personalized continuous theta-burst stimulation (cTBS) targeting the amygdala and its circuitry may be a potential therapeutic approach for this population.
Methods: In a double-blind randomized controlled trial, minimally verbal children with autism (ages 2-8 years) received 4 weeks of cTBS.

Membrane potential (MP) changes can provide a simple readout of bacterial functional and metabolic state or stress levels. While several optical methods exist for measuring fast changes in MP in excitable cells, there is a dearth of such methods for absolute and precise measurements of steady-state membrane potentials (MPs) in bacterial cells. Conventional electrode-based methods for the measurement of MP are not suitable for calibrating optical methods in small bacterial cells.

Background: Opioid use disorder remains a critical healthcare challenge as current therapeutic strategies have limitations resulting in high recurrence and deaths. We evaluated safety and feasibility of focused ultrasound (FUS) neuromodulation to reduce substance cravings and use in severe opioid- and co-occurring substance use disorders.
Methods: This prospective, open-label, single-arm study enrolled 8 participants with severe, primary opioid use disorder with co-occurring substance use.

Enhancing the retention of recent memory traces through sleep reactivation is possible via Targeted Memory Reactivation (TMR), involving cueing learned material during post-training sleep. Evidence indicates detectable short-term microstructural changes in the brain within an hour after motor sequence learning, and post-training sleep is believed to contribute to the consolidation of these motor memories, potentially leading to enduring microstructural changes. In this study, we explored how TMR during post-training sleep affects performance gains and delayed microstructural remodeling, using both standard Diffusion Tensor Imaging (DTI) and advanced Neurite Orientation Dispersion & Density Imaging (NODDI).

Norepinephrine in vertebrates and its invertebrate analog, octopamine, regulate the activity of neural circuits. We find that, when hungry, larvae switch activity in type II octopaminergic motor neurons (MNs) to high-frequency bursts, which coincide with locomotion-driving bursts in type I glutamatergic MNs that converge on the same muscles. Optical quantal analysis across hundreds of synapses simultaneously reveals that octopamine potentiates glutamate release by tonic type Ib MNs, but not phasic type Is MNs, and occurs via the G-coupled octopamine receptor (OAMB).

Dysfunctional response inhibition, mediated by the striatum and its connections, is thought to underly the clinical manifestations of obsessive-compulsive disorder (OCD). However, the exact neural mechanisms remain controversial. In this study, we undertook a novel approach by positing that a) inhibition is a dynamic construct inherently susceptible to numerous failures, which require error-processing, and b) the actor-critic framework of reinforcement learning can integrate neural patterns of inhibition and error-processing in OCD with their behavioural correlates.

This paper introduces the Automated Lesion and Feature Extraction (ALFE) pipeline, an open-source, Python-based pipeline that consumes MR images of the brain and produces anatomical segmentations, lesion segmentations, and human-interpretable imaging features describing the lesions in the brain. ALFE pipeline is modeled after the neuroradiology workflow and generates features that can be used by physicians for quantitative analysis of clinical brain MRIs and for machine learning applications. The pipeline uses a decoupled design which allows the user to customize the image processing, image registrations, and AI segmentation tools without the need to change the business logic of the pipeline.

Neurons in the hippocampus are correlated with different variables, including space, time, sensory cues, rewards and actions, in which the extent of tuning depends on ongoing task demands. Here we present a rare case of hemorrhagic longitudinally extensive transverse myelitis (LETM) as a first presentation of Neuromyelitis Optica Spectrum Disorder. Even though the patient received aggressive treatment, he showed no significant improvement. Our case highlights the importance of prompt intervention in the case of hemorrhagic myelitis and the diagnostic challenges of hemorrhagic LETM.

Schemas are rich and complex knowledge structures about the typical unfolding of events in a context; for example, a schema of a dinner at a restaurant. In this Perspective, we suggest that reinforcement learning (RL), a computational theory of learning the structure of the world and relevant goal-oriented behaviour, underlies schema learning. We synthesize literature about schemas and RL to offer that three RL principles might govern the learning of schemas: learning via prediction errors, constructing hierarchical knowledge using hierarchical RL, and dimensionality reduction through learning a simplified and abstract representation of the world.
