Detection of bacterial gene expression in soil emerged in the early 1990s and provided information on bacterial responses in their original soil environments. As a key procedure in the detection, extraction of bacterial RNA from soil has attracted much interest, and many methods of soil RNA extraction have been reported in the past 20 years. In addition to various RT-PCR-based technologies, new technologies for gene expression analysis, such as microarrays and high-throughput sequencing technologies, have recently been applied to examine bacterial gene expression in soil.

Pseudozyma antarctica JCM 10317 exhibits a strong degradation activity for biodegradable plastics (BPs) such as agricultural mulch films composed of poly(butylene succinate) (PBS) and poly(butylene succinate-co-adipate) (PBSA). An enzyme named PaE was isolated and the gene encoding PaE was cloned from the strain by functional complementation in Saccharomyces cerevisiae. The deduced amino acid sequence of PaE contains 198 amino acids with a predicted molecular weight of 20,362.

The main objective of this paper was to assess the suitability of an enzyme-linked immunosorbent assay (ELISA) as a rapid and simple screening method for the detection of neonicotinoid insecticide dinotefuran residues in various vegetable samples including carrot, cabbage, green pepper, leek, Japanese mustard spinach (komatsuna), and spinach. Simple dilution of crude sample extracts was effective to circumvent or reduce matrix interference derived from samples. Consequently, ELISA was useful to determine dinotefuran accurately and directly, only requiring methanol extraction and dilution with water.

Rapid evolution has led to a large diversity in the sizes and morphology of male genitals across taxa, but the mechanisms driving this evolution remain controversial. In this study, we investigated the function of male genital sclerites in the adzuki bean beetle (Callosobruchus chinensis) and compared the length and morphology of genital sclerites between two populations that vary in their degree of polyandry. We found that the length of male genital sclerites was negatively correlated with copulation duration but positively correlated with the speed of matings with multiple females.

Three weeks after the accident at the Fukushima Daiichi Nuclear Power Plant, we determined the activity concentrations of (131)I, (134)Cs and (137)Cs in atmospheric dust fugitively resuspended from soil particles due to soil surface perturbation by agricultural practices. The atmospheric concentrations of (131)I, (134)Cs and (137)Cs increased because of the agitation of soil particles by a hammer-knife mower and a rotary tiller. Coarse soil particles were primarily agitated by the perturbation of the soil surface of Andosols.

The specificity of culturable bacteria on healthy and Fusarium head blight (FHB)-infected spikelets of wheat heads was investigated to find a candidate of biocontrol agents against FHB. The bacterial genus Pseudomonas was commonly isolated from the tissues, and phylogenetic analysis using 16S ribosomal RNA gene sequences of isolates of the genera revealed that particular phylogenetic groups in the genus specifically inhabited either healthy or infected spikelet tissues. The specificity of each group was suggested to be due to differences in the ability to form biofilms and colonize spikelet tissues; isolates originated from healthy spikelets formed biofilms on polyvinyl chloride microplate wells and highly colonized the spikelet tissues.

We developed a dataset of local-scale daily climate change scenarios for Japan (called ELPIS-JP) using the stochastic weather generators (WGs) LARS-WG and, in part, WXGEN. The ELPIS-JP dataset is based on the observed (or estimated) daily weather data for seven climatic variables (daily mean, maximum and minimum temperatures; precipitation; solar radiation; relative humidity; and wind speed) at 938 sites in Japan and climate projections from the multi-model ensemble of global climate models (GCMs) used in the coupled model intercomparison project (CMIP3) and multi-model ensemble of regional climate models form the Japanese downscaling project (called S-5-3). The capability of the WGs to reproduce the statistical features of the observed data for the period 1981-2000 is assessed using several statistical tests and quantile-quantile plots.

Heavy metals are transported to rice grains via the phloem. In rice nodes, the diffuse vascular bundles (DVBs), which enclose the enlarged elliptical vascular bundles (EVBs), are connected to the panicle and have a morphological feature that facilitates xylem-to-phloem transfer. To find a mechanism for restricting cadmium (Cd) transport into grains, the distribution of Cd, zinc (Zn), manganese (Mn), and sulphur (S) around the vascular bundles in node I (the node beneath the panicle) of Oryza sativa 'Koshihikari' were compared 1 week after heading.
View Article and Find Full Text PDFWe simultaneously examined the bacteria, fungi and nematode communities in Andosols from four agro-geographical sites in Japan using polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) and statistical analyses to test the effects of environmental factors including soil properties on these communities depending on geographical sites. Statistical analyses such as Principal component analysis (PCA) and Redundancy analysis (RDA) revealed that the compositions of the three soil biota communities were strongly affected by geographical sites, which were in turn strongly associated with soil characteristics such as total C (TC), total N (TN), C/N ratio and annual mean soil temperature (ST). In particular, the TC, TN and C/N ratio had stronger effects on bacterial and fungal communities than on the nematode community.

Population cycles have fascinated ecologists since the early nineteenth century, and the dynamics of insect populations have been central to understanding the intrinsic and extrinsic biological processes responsible for these cycles. We analyzed an extraordinary long-term data set (every 5 days for 48 years) of a tea tortrix moth (Adoxophyes honmai) that exhibits two dominant cycles: an annual cycle with a conspicuous pattern of four or five single-generation cycles superimposed on it. General theory offers several candidate mechanisms for generation cycles.

Ammonia monooxygenase subunit A gene (amoA) is frequently used as a functional gene marker for diversity analysis of ammonia-oxidizing bacteria (AOB). To select a suitable amoA primer for real-time PCR and PCR-denaturing gradient gel electrophoresis (DGGE), three reverse primers (degenerate primer amoA-2R; non-degenerate primers amoA-2R-GG and amoA-2IR) were examined. No significant differences were observed among the three primers in terms of quantitative values of amoA from environmental samples using real-time PCR.
