Vaccination is the most effective method used to reduce the morbidity and mortality of influenza infections. However, as exemplified in the current swine-origin influenza virus (S-OIV) pandemic, the global manufacturing capacity of influenza vaccines is severely limited. In the present proof-of-concept study, we combined cell substrate selection and antigen engineering with adjuvant development to design a potential pandemic influenza vaccine candidate, in which CpG oligodeoxynucleotides (CpG-ODN) plus alum was used as a composite adjuvant to enhance the immunogenicity of insect cell-expressed recombinant hemagglutinin (rHA).

Objective: To establish a quantitative assay for enterovirus 71, this can be used in detecting the virus content during vaccine development and production.

Methods: We established the method of quantitative assay for EV71 by using double antibody sandwich ELISA. The sensitivity, accuracy, precision and specificity of the method were evaluated.

Poliovirus transmission is controlled globally through world-wide use of a live attenuated oral polio vaccine (OPV). However, the imminence of global poliovirus eradication calls for a switch to the inactivated polio vaccine (IPV). Given the limited manufacturing capacity and high cost of IPV, this switch is unlikely in most developing and undeveloped countries.

Objective: To establish and optimize methods to detect the immune complexes (IC) of hepatitis B virus directly.

Methods: A C1q solid phase ELISA, mouse anti-HBs MAb solid phase ELISA and the complement consumption assay were established to detect the IC and these methods were optimized.

Results: All the three methods were highly sensitive, specific and reproducible.

Corynebacterium diphtheria PW8-weissensee was grown on different media for screening high toxin produce. Through six times of screening (selecting butyrous brilliant smooth small colonies and testing their toxin production capacity) from original strain producing toxin 160 Lf/ml, a high toxin--producing descendand culture was obtained, which produced 79% more toxin than the original strain did.

Objective: To study immunogenecity of yeast-derived recombinant hepatitis B (YHB) component in diphtheria-pertussis-tetanus-YHB vaccine (DPTw-YHB).

Methods: Immunogenecity of tetra-valent DTPw-YHB vaccine and mono-valent recombinant YHB vaccine, and that of the tetra-valent vaccine with varied YHB component were compared. The efficiency and stability of recombinant YHB in the tetra-valent vaccine stored at 2 - 8 degrees C for 18 months were determined.

[Studies on the molecular epidemiology of poliovirus type 1 in China].

Zhonghua Liu Xing Bing Xue Za Zhi

August 1997

Sino-Danish Biomedical Postgraduate Training Center, National Vaccine and Serum Institute, Beijing.

A strain of poliovirus type 1 isolated from an endemic in Shandong Province in 1988 and three strains of poliovirus type 1 isolated from an endemic in Xinjiang in 1990 were analysed by dot blot hybridization with Sabin I specific probe and PCR amplification with Sabin I specific primers. They were proved to be non-Sabin-like poliovirus. The nucleotide sequences of VP1-2A junction region of four poliovirus type 1 isolates were analysed.

The Sf9 cell line, commonly used for gene expression by recombinant baculoviruses, can be productively infected by Japanese B encephalitis virus (JEV). Two wild-type JEV strains (P3 and SA14) caused a cytopathic effect (CPE) in the Sf9 cells, while no apparent CPE was caused by an attenuated strain (SA14-14-2). The JEV viral antigens were expressed in the infected Sf9 cells and intracellular virus particles were found by electron microscopy as a result of infection with all three strains.

In order to evaluate the difference between plaque reduction neutralization test (PNt) and hemagglutination inhibition (HI) test on human anti-measles antibodies, pre- and postimmunization sera collected from 328 infants aged 3-6 months who were immunized with Shanghai 191 measles vaccine, were detected by both methods. The results demonstrated that PNt was more sensitive than HI when they were used to measure the pre-immunization sera. No significant difference was found between these two methods when they were used to measure the post-immunization sera.

A series of trachoma inclusion color photograph is taken from the conjunctival smears of 48 trachoma cases. These smears were Giemsa-stained and stored in the slide-boxes for more than 30 years with no distinct changes in color. A description is given to the Pleomorphism of trachoma inclusion and its relation to the host cells.

A comparison study on the responses to the immunization of three different forms of polio vaccine, eg. Liquid I, Oral dragee candy I and Trivalent Oral dragee candy, on the susceptible children of 2-6 month of age was conducted in wujin County, Jiangsu Province The study showed no statistical difference between the responses to the three forms of the vaccine reaching 95-100%. For antibody GMT, Liquid I was the highest, with little difference between Liquid I and Oral dragee candy I and Trivalent Oral dragee candy vaccine in statistics.

This artical reports the result of serological observation on an improved immunization schedule of polio vaccine. The result shows that a good response can be obtained by immunizing an additional dose of the vaccine on the children of 18-24 month of age.serological convertion type I of neutralization antibody can be improved from 85% up to 96%, type II from 84% to 100%, and type III from 91% to 100%, with great increase of all the three types.
