Taking advantage of the good mechanical strength of expanded Drosophila brains and to tackle their relatively large size that can complicate imaging, we apply potassium (poly)acrylate-based hydrogels for expansion microscopy (ExM), resulting in a 40x plus increased resolution of transgenic fluorescent proteins preserved by glutaraldehyde fixation in the nervous system. Large-volume ExM is realized by using an axicon-based Bessel lightsheet microscope, featuring gentle multi-color fluorophore excitation and intrinsic optical sectioning capability, enabling visualization of Tm5a neurites and L3 lamina neurons with photoreceptors in the optic lobe. We also image nanometer-sized dopaminergic neurons across the same intact iteratively expanded Drosophila brain, enabling us to measure the 3D expansion ratio.

Steering acidic oxygen reduction selectivity of single-atom catalysts through the second sphere effect.

Natural enzymes feature distinctive second spheres near their active sites, leading to exquisite catalytic reactivity. However, incumbent synthetic strategies offer limited versatility in functionalizing the second spheres of heterogeneous catalysts. Here, we prepare an enzyme-mimetic single Co-N atom catalyst with an elaborately configured pendant amine group in the second sphere via 1,3-dipolar cycloaddition, which switches the oxygen reduction reaction selectivity from the 4e to the 2e pathway under acidic conditions.

Immune checkpoint inhibitors (ICI) represent new anticancer agents and have been used worldwide. However, ICI can potentially induce life-threatening severe cutaneous adverse reaction (SCAR), such as Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN), hindering continuous ICI therapy. We examine 6 cohorts including 25 ICI-induced SJS/TEN patients and conduct single-cell RNA sequencing (scRNA-seq) analysis, which shows overexpression of macrophage-derived CXCL10 that recruits CXCR3 cytotoxic T lymphocytes (CTL) in blister cells from ICI-SJS/TEN skin lesions.

However, existing methods used to assess autophagy activity often pose invasiveness and time-related constraints, limiting their applicability in preclinical drug investigations. In this study, we developed a non-invasive autophagy detection system (NIADS-autophagy, also called G-cleave LC3B biosensor) by integrating a split-luciferase-based biosensor with an LC3B cleavage sequence, which swiftly identified classic autophagic triggers, such as Earle's Balanced Salt Solution and serum deprivation, through protease-mediated degradation pathways.

Bilateral inverse canine impaction: a case report.

Background: Maxillary canine impaction can result in malocclusion, temporomandibular joint complications, and esthetic concerns, thus influencing craniofacial development. Although cases of impacted canines are not uncommon in clinical practice, the likelihood of simultaneous bilateral impaction is extremely low; very few reports have documented cases of bilaterally symmetrical, inversely impacted canines.

The purpose of this study was to investigate the effects of acetyl tributyl citrate (ATBC) on the mechanical properties, abrasion resistance, and cytotoxicity of a polyurethane-based 3D printing resin for mouthguard applications. The synthesized polycarbonate-based polyurethane acrylate was formulated into digital light processing printing resins with 40 wt% triethylene glycol dimethacrylate, and different percentage of ATBC were added for further characterizations. The mechanical properties and abrasion resistance, ATBC migration, and the cytotoxicity of the resins were evaluated.
