Human peripheral blood lymphocytes (PBLs) produce metallothioneins (MTs) in response to a variety of heavy metal ions. MTs could therefore be a candidate for a marker that represents the biological effect of heavy metals. Cyclohexane was single out as a candidate for the alternative vapor out of six vapors: trichloroethylene, n-pentane, n-hexane, n-heptane, toluene and cyclohexane from the viewpoints of similarity of vapor pressure and water-solubility to CCl4, lower toxicity than CCl4, and technical feasibility in generating an airflow of 30 L/min containing the vapor ranging from 300 to 5000 ppm. Breakthrough times of cartridges and canisters were examined by generating cyclohexane vapor in a test airflow of 30 L/min in comparison with CCl4 vapor under various test conditions with different vapor concentrations and relative humidities (RH).

View Article and Find Full Text PDFIn order to improve arc welding work in a small enclosed workspace, numerical simulations were conducted to find the most appropriate welding and ventilation conditions, such as welding currents, hood position and flow rates with no blowhole formation. In the simulations, distributions of airflow vectors and fume concentrations were calculated for two hood opening positions: one faced a welder's breathing zone, the other a contaminant source. As a result it was predicted that a hood opening facing a breathing zone remarkably lowered the fume concentration in the breathing zone compared with that facing a contaminant source.

Mammalian metallothionein genes are transcriptionally regulated by heavy metals through cis-acting metal responsive elements (MREs). The MRE-binding transcription factor-1 (MTF-1), a protein containing six C(2)H(2)-type Zn fingers, is essential for MRE-mediated transcriptional activation. DNA binding of MTF-1 is known to be stimulated by Zn in vitro, but the binding was also largely influenced by redox conditions, suggesting that redox signals could modulate MTF-1 activity.

We examined the effects of a 15-min nap after lunch on subsequent alertness, performance, and autonomic function following a short sleep the preceding night. Subjects were 12 healthy students who had slept for only 4 hours the night before being tested. They experienced both nap and no-nap conditions in a counterbalanced order, at least a week apart.

Changes in the binding affinity of the muscarinic acetylcholine receptor agonist carbamylcholine were determined in membranes isolated from the brains of rats exposed to toluene at concentrations of 500-2,000 ppm for 6 h. Membrane fractions of the frontal cortex and hippocampus were prepared and agonist-binding affinities were determined by measuring the displacement of [3H]N-methyl scopolamine-binding activity by carbamylcholine. In the frontal cortex, the affinity of high-affinity carbamylcholine binding was reduced following exposure to toluene at a concentration of 1000 ppm or higher.

The exposure system allowed an inhalation of fresh welding fumes and gases, and could supply airflow containing stable concentrations of fumes and ozone even the levels exceeding those corresponding occupational exposure limit values were supplied into the exposure chamber. The air temperature in the chamber was kept constant under rat's physiological conditions.

Heavy metals are expected to affect the biological activity of various metal-containing proteins, including transcriptional regulators. We studied the effects of several heavy metal ions on the DNA-binding activity of a Zn-finger transcription factor, Sp1. With respect to both DNA elements through which Sp1 acts positively and negatively, Cd2+ inhibited DNA-binding of Sp1 at 20 microM or higher, while the toxic effect of Zn2+ was obvious only at more than 200 microM.

The occupational exposure limit for acrylonitrile (AN) has been set by many organizations on the basis of its carcinogenicity. However, recent epidemiological studies do not afford evidence supporting the hypothesis that AN is carcinogenic to humans. Review of the 18 published cohort studies revealed that, although there is not adequate evidence in humans for carcinogenicity of AN, the possibility of a causal association between high exposure to AN and lung cancer in humans cannot be excluded.
View Article and Find Full Text PDFTo examine the possible roles of mast cells in the pathogenesis of subcapsular cell hyperplasia (SCH) in the adrenal glands of mice, we investigated the expression of certain cytokines, including stem cell factor (SCF), tumor necrosis factor-alpha (TNF-alpha), nerve growth factor (NGF), and basic fibroblast growth factor (bFGF), and mast cell-specific proteases, such as mouse mast cell protease (mMCP)-2 and mMCP-7. The mRNAs of c-kit (SCF receptor), bFGF, TNF-alpha, mMCP-2, and mMCP-7 were expressed in both the adrenal glands and the mouse bone marrow-derived mast cells (mBMMCs). Immunoreactivities for cytokines (SCF, NGF, TNF-alpha) and proteases (mMCP-2, mMCP-7) were exclusively located in the mast cells in SCH lesions.

To examine how repeated cooling of fingers with a rest pause schedule at work affects cold-induced vasodilatation (CIVD), pain and cold sensation in fingers, six healthy men aged 21 to 23 years immersed their left index fingers six times in stirred water at 10 degrees C for 10 minutes. After each cold-water immersion of the fingers, 5-minute rest pause was taken to observe the recovery process of the indicators. This cold-water immersion/rest pause test was carried out in a range of three ambient temperature conditions: 30 degrees C (warm), 25 degrees C (thermoneutral), and 20 degrees C (cool) as experienced in daily life.
View Article and Find Full Text PDFConcentrations of fumes, ozone (O3), carbon monoxide (CO), nitric oxide (NO), manganese (Mn) and total and hexavalent chromium (Cr) as well as size distribution of fumes were measured at a point corresponding to the welder's breathing zone during CO2-arc welding, using a welding robot and three kinds of wires. Concentrations of fumes, O3, CO, Mn and total-Cr were found to exceed their corresponding occupational exposure limit (OEL) values, while the concentrations of NO and Cr(VI) were below those OEL levels. Airborne concentration of Mn exceeded its OEL value, and the Mn content was 8 times higher in welding fumes than in the wire.

In order to clarify poor sleep habits and prevalence in sleep problems (disturbances) of healthy male white-collar workers and the relationship of these to age and job type, a total of 1,161 Japanese employees of an electric equipment manufacturing company (aged 23 to 59, mean 37 years) were surveyed by means of a mailed questionnaire. The workers were asked about eleven sleep habits. They were also divided into four groups according to age and job type: 23-29 (n = 226), 30-39 (n = 597), 40-49 (n = 225) and 50-59 (n = 113); managerial (n = 209), professional (n = 336), technical (n = 475) and clerical (n = 134).

We determined the effects of toluene exposure on activation or inhibition of adenylyl cyclase by stimulating human beta2-adrenergic receptors (beta2-AR) and muscarinic acetylcholine receptor (mAChR) m2 subtypes, respectively, expressed in CHO cells. The objective of the present study was to determine perception thresholds for whole-body vibration in the recumbent supine posture in vertical and horizontal directions and to compare the present results with ones obtained at the same laboratory in 1984 and also with ISO frequency weighting curves both in ISO 2631-1 and in ISO 2631-2. The frequency characteristic and thresholds values of the present results are similar to previous results. Weighting curves of ISO, however, show quite different characteristic in both vertical and horizontal directions compared to the present characteristics.
