The applications of resveratrol (RES) and puerarin (PUE) with notable physiological functions are greatly limited in functional food and pharmaceutical industries due to their poor water solubility and chemical instability. Chronic exposure of the liver to multiple insults culminates in the development of metabolic dysfunctionassociated steatohepatitis (MASH), a complicated metabolic syndrome characterized by hepatic steatosis and inflammation, typically accompanied by progressive fibrosis. Despite extensive clinical evaluation, there remain challenges in MASH drug development, which are primarily due to unsatisfactory efficacy and limited specificity. Strategies to address the unmet medical need for MASH with fibrosis before it reaches the irreversible stage of decompensated cirrhosis are critically needed.

Cancer, a key factor in declining global life expectancy, has driven the integration of chemotherapy and immunotherapy to address multidrug resistance and influence the tumor microenvironment. We developed a novel vaccine delivery carrier, a chitosan-coated polylactic acid/poloxamer nanoparticle (CPP NP), designed to co-encapsulate an anticancer drug and antigen without any chemical conjugation process, enabling effective and synergistic cancer chemo-immunotherapy. The CPP NP achieved synergistic efficacy through paclitaxel (PTX), an immunogenic cell death-inducing chemotherapeutic agent; ovalbumin (OVA), which promotes dendritic cell maturation; and enhanced cellular uptake facilitated by chitosan.

Delivering ribonucleoproteins (RNPs) for in vivo genome editing is safer than using viruses encoding for Cas9 and its respective guide RNA. However, transient RNP activity does not typically lead to optimal editing outcomes. Here we show that the efficiency of delivering RNPs can be enhanced by cell-penetrating peptides (covalently fused to the protein or as excipients) and that lipid nanoparticles (LNPs) encapsulating RNPs can be optimized for enhanced RNP stability, delivery efficiency and editing potency.

A practical and efficient method for the -acrylation of amines with (trifluoromethyl)alkenes is achieved the cleavage of three C(sp)-F bonds, affording a diverse range of useful tertiary and secondary α-arylacrylamides in high yields. This protocol features mild conditions, is transition-metal free, operationally simple, gram-scalable, and compatible with valuable functional groups, and has a broad substrate scope. Mechanistic studies indicate that exchange of an oxygen atom happens between HO and NaOH, and that the oxygen atom is incorporated into the α-arylacrylamides the -defluorooxylation of the (trifluoromethyl)alkene.
