Motivation: Spaln is the earliest practical tool for self-sufficient genome mapping and spliced alignment of protein query sequences onto a mammalian-sized eukaryotic genomic sequence. However, its computational speed has become inadequate for the analysis of rapidly growing genomic and transcript sequence data.
Results: The dynamic programming calculation of Spaln has been sped up in two ways: (i) the introduction of the multi-intermediate unidirectional Hirschberg method and (ii) SIMD-based vectorization.

We genetically manipulated HaCaT cells, a spontaneously immortalised normal keratinocyte cell line, to stably express two different coloured luciferase reporter genes, driven by interleukin 8 (IL-8) and ubiquitin-C (UBC) promoters, respectively. Subsequently, we generated a three-dimensional (3D) skin-like in vitro composite (SLIC) utilising these cells, with the objective of monitoring bioluminescence emitted from the SLIC. This SLIC was generated on non-woven silica fibre membranes in differentiation medium.

Generally, activated carbons demonstrated a notable ability to capture long-chain PFAS, but exhibited relatively lower effectiveness for short-chain PFAS. Thirteen commercially available activated carbons in Japan underwent testing for their adsorption capacity of PFAS in water. The activated carbon derived from rice husk, Triporous™-PFAS, exhibited the highest adsorption capacity (over 95%) for PFAS from ultrashort-chain (perfluorocarbon chain: C1 for perfluorocarboxylic acid (PFCA) and C2 for perfluoroalkane sulfonic acid (PFSA)) to long-chain PFAS (C13 for PFCA and C10 for PFSA).

Bioluminescence (BL) generated by luciferase-coelenterazine (CTZ) reactions is broadly employed as an optical readout in bioassays and in vivo molecular imaging. In this study, we demonstrate a systematic approach to elucidate the luciferase-CTZ binding chemistry with a full set of regioisomeric CTZ analogs, where all the functional groups were regiochemically modified. When the chemical structures were categorized into Groups 1-6, the even-numbered Groups (2, 4, and 6) of the CTZ analogs are found to be exceptionally bright with NanoLuc enzyme.

We have investigated the in-plane local structure of the BaKFeAs superconductor by polarized Fe K-edge extended X-ray absorption fine structure (EXAFS) measurements with temperature. The near neighbor bond distances and their stiffness, measured by polarized EXAFS in two orthogonal directions, are different suggesting in-plane anisotropy of the atomic displacements and local orthorhombicity in the title system. The X-ray absorption near edge structure (XANES) spectra reveal anisotropy of valence electronic structure that changes anomalously below ∼100 K.

The genome-editing efficiency of the CRISPR-Cas9 system hinges on the recognition of the protospacer adjacent motif (PAM) sequence, which is essential for Cas9 binding to DNA. The commonly used (SpyCas9) targets the 5'-NGG-3' PAM sequence, which does not cover all the potential genomic-editing sites. To expand the toolbox for genome editing, SpyCas9 has been engineered to recognize flexible PAM sequences and Cas9 orthologs have been used to recognize novel PAM sequences.

Nanostructured lead telluride PbTe is among the best-performing thermoelectric materials, for both p- and n-types, for intermediate temperature applications. However, the fabrication of power-generating modules based on nanostructured PbTe still faces challenges related to the stability of the materials, especially nanoprecipitates, and the bonding of electric contacts. In this study, in situ high-temperature transmission electron microscopy observation confirmed the stability of nanoprecipitates in p-type PbNaGeTe up to at least ∼786 K.
