Dextran-based nanodrugs with mitochondrial targeting/glutathione depleting synergy for enhanced photodynamic therapy.

Carbohydr Polym

State Key Laboratory of Metal Matrix Composites, School of Materials Science and Engineering, Shanghai Jiao Tong University, Shanghai 200240, China. Electronic address:

The efficacy of photodynamic therapy (PDT) for malignant tumors is significantly impeded by the short diffusion distance of reactive oxygen species (ROS) and the ROS-consuming glutathione (GSH) overexpressed in tumor cells. Therefore, enhanced PDT can be achieved by the construction of biomacromolecule-based nanodrugs that can specifically target ROS-sensitive mitochondria and deplete intracellular GSH. Herein, we synthesized the dextran-based nano-assemblies by a Graft copolymerization Induced Self-Assembly (GISA) method, in which methyl acrylate and diallyl disulfide (DADS) were copolymerized from a mixed dextran/amino dextran backbone in an aqueous medium.

Macromolecular Nano-Assemblies for Enhancing the Effect of Oxygen-Dependent Photodynamic Therapy Against Hypoxic Tumors.

Chemistry

August 2024

State Key Laboratory of Metal Matrix Composites, School of Materials Science and Engineering, Shanghai Jiao Tong University, 800 Dongchuan Road, 200240, Shanghai, China.

In oxygen (O)-dependent photodynamic therapy (PDT), photosensitizers absorb light energy, which is then transferred to ambient O and subsequently generates cytotoxic singlet oxygen (O). Therefore, the availability of O and the utilization efficiency of generated O are two significant factors that influence the effectiveness of PDT. However, tumor microenvironments (TMEs) characterized by hypoxia and limited utilization efficiency of O resulting from its short half-life and short diffusion distance significantly restrict the applicability of PDT for hypoxic tumors.

Biological samples are important resources for scientific research. These samples are stored in biobanks over years until needed, and some of them can never be retrieved if they are improperly stored, causing them to be wasted. Thus, they are priceless, and they should be used correctly and effectively.

Single-Cell Diagnosis of Cancer Drug Resistance through the Differential Endocytosis of Nanoparticles between Drug-Resistant and Drug-Sensitive Cancer Cells.

ACS Nano

October 2023

The State Key Laboratory of Metal Matrix Composites, School of Materials Science and Engineering, Shanghai Jiao Tong University, 800 Dongchuan Road, Shanghai 200240, China.

Single-cell diagnosis of cancer drug resistance is highly relevant for cancer treatment, as it can be used to identify the subpopulations of drug-resistant cancer cells, reveal the sensitivity of cancer cells to treatment, and monitor the progress of cancer drug resistance. However, simple and effective methods for cancer drug resistance detection at the single-cell level are still lacking in laboratory and clinical studies. Inspired by the fact that nanoparticles with diverse physicochemical properties would generate distinct and specific interactions with drug-resistant and drug-sensitive cancer cells, which have distinctive molecular signatures, here, we have synthesized a library of fluorescent nanoparticles with various sizes, surface charges, and compositions (SiO nanoparticles (SNPs), organic PS--PAA nanoparticles (ONPs), and ZIF-8 nanoparticles (ZNPs)), thus demonstrating that the composition has a critical influence on the interaction of nanoparticles with drug-resistant cancer cells.

Cryopreservation and Transplantation of Spermatogonial Stem Cells.

Methods Mol Biol

March 2021

Department of Aquaculture, Szent István University, Gödöllő, Páter Károly u. 1., Hungary.

Cryopreservation as a method that enables long-term storage of biological material has long been used for the conservation of valuable zebrafish genetic resources. However, currently, only spermatozoa of zebrafish can be successfully cryopreserved, while protocols for cryopreservation of eggs and embryos have not yet been fully developed. Transplantation of germline stem cells (GSCs) has risen as a favorable method that can bypass the current problem in cryopreservation of female genetic resources and can lead to reconstitution of fish species and lines through surrogate production.
