J Autoimmun

March 2025

National Center for Clinical Laboratories, Institute of Geriatric Medicine, Chinese Academy of Medical Sciences, Beijing Hospital/National Center of Gerontology, Beijing, 100730, China; Department of Rheumatology, Beijing Hospital, National Center of Gerontology, Institute of Geriatric Medicine, Clinical Immunology Center, Chinese Academy of Medical Sciences and Peking Union Medical College, Beijing, China. Electronic address:

Objective: Aberrant neutrophil activation is implicated in the pathogenesis of systemic lupus erythematosus (SLE) and its related comorbidities. We found that CD177 was one of the most highly up-regulated genes at the transcriptional level in purified neutrophils from SLE patients. In this study, we aimed to explore the role of CD177 neutrophils in the pathogenesis of SLE.

Objectives: We evaluated the intermediate reproducibility imprecision of cystatin C results based on internal quality control (IQC) data.

Methods: Five performance specifications [1/3 total allowable error (TEa), 1/4TEa and three levels performance specifications based on biological variation] were used to calculate the proportion of laboratories with CVs less than or equal to the performance specifications, namely the pass rate. Based on the reference interval of Chinese adult serum cystatin C (0.

Objectives: To improve the accuracy of lymphocyte subsets counts, a traceable dual-platform extended PanLeucogating method (DPP) of absolute cell counts of lymphocyte subsets was introduced, and consistency was evaluated by comparing conventional single/dual-platform method with DPP.

Methods: The DPP for absolute lymphocyte subsets counts was established by multiplying the percentage of lymphocyte subsets in total white blood cells (WBC) measured by flow cytometer with the total WBC counts. DPP-R was defined as the use of the total WBC counts measured in reference laboratory that was traceable to reference method recommended by International Council for Standardization in Hematology (ICSH).

Construction of RNA reference materials for improving the quantification of transcriptomic data.

Nat Protoc

February 2025

State Key Laboratory of Genetic Engineering, Human Phenome Institute and School of Life Sciences, Fudan University, Shanghai, China.

RNA reference materials and their corresponding reference datasets act as the 'ground truth' for the normalization of experimental values and are indispensable tools for reliably measuring intrinsically small differences in RNA-sequencing data, such as those between molecular subtypes of diseases in clinical samples. However, the variability in 'absolute' expression profiles measured across different batches, methods or platforms limits the use of conventional RNA reference datasets. We recently proposed a ratio-based method for constructing reference datasets.

Analytical performance evaluation and optimization of serum 25(OH)D LC-MS/MS measurement.

Clin Chem Lab Med

February 2025

National Center for Clinical Laboratories, Institute of Geriatric Medicine, Chinese Academy of Medical Sciences, Beijing Hospital/ National Center of Gerontology, Beijing, China.

Objectives: Measuring serum 25-hydroxyvitamin D is key in clinical labs, but inter-laboratory variations risk diagnostic errors. Objectives: Little is known about the diversity of residents' preferences for COVID-19 vaccines during the time when COVID-19 management was downgraded in China. This study aims to investigate these preferences using discrete choice experiment (DCE) and profile case best-worst scaling (BWS-2), and to assess the concordance between these two methods.

Methods: Chinese residents recruited for the online survey were asked to evaluate COVID-19 vaccine profiles through both DCE and BWS-2 from April to July 2023.

Humans are widely exposed to phenols, parabens, and phthalates with health risks, while the effects of these chemicals on biological aging remain unclear. Among 3,441 adults in the National Health and Nutrition Examination Survey 2005-2010, phenol, paraben, and phthalate concentrations were measured and phenotypic age acceleration (PhenoAgeAccel) was calculated. Linear regression and weighted quantile sum (WQS) regression were used to evaluate the associations of single and mixed chemicals with PhenoAgeAccel.

Interlaboratory consistency of promoter methylation detection in colorectal cancer using the post-optimized materials.

iScience

November 2024

National Center for Clinical Laboratories, Institute of Geriatric Medicine, Chinese Academy of Medical Sciences, Beijing Hospital/ National Center of Gerontology, Beijing, P.R. China.

Fecal DNA-based Syndecan 2 () methylation detection is a promising non-invasive strategy for early colorectal cancer (CRC) screening. In China, commercial assays for methylation detection vary in sensitivity and specificity, yet there is no standardized external quality assessment (EQA) to ensure accuracy. This study utilized CRISPR-Cas9 and homology-directed repair (HDR) technologies to edit the promoter in 293T cells, creating hypermethylated and heterogeneous cell lines.
