Bacterial peptidoglycan, the essential cell surface polymer that protects bacterial integrity, also serves as the molecular pattern recognized by the host's innate immune system. Although the minimal motifs of bacterial peptidoglycan fragments (PGNs) that activate mammalian NOD1 and NOD2 sensors are well-known and often represented by small canonical ligands, the immunostimulatory effects of natural PGNs, which are structurally more complex and potentially can simultaneously activate both the NOD1 and NOD2 signaling pathways in hosts, have not been comprehensively investigated. In particular, many bacteria incorporate additional structural modifications in peptidoglycans to evade host immune surveillance, resulting in diverse structural variations among natural PGNs that may influence their biological effects in hosts.

Hepatocellular carcinoma (HCC) is accompanied by an altered gut microbiota but whether the latter contributes to carcinogenesis is unclear. Here we show that faecal microbiota transplantation (FMT) using stool samples from patients with HCC spontaneously initiate liver inflammation, fibrosis and dysplasia in wild-type mice, and accelerate disease progression in a mouse model of HCC. We find that HCC-FMT results in gut barrier injury and translocation of live bacteria to the liver.

Hardware implementation of reconfigurable and nonvolatile photoresponsivity is essential for advancing in-sensor computing for machine vision applications. However, existing reconfigurable photoresponsivity essentially depends on the photovoltaic effect of p-n junctions, which photoelectric efficiency is constrained by Shockley-Queisser limit and hinders the achievement of high-performance nonvolatile photoresponsivity. Here, we employ bulk photovoltaic effect of rhombohedral (3R) stacked/interlayer sliding tungsten disulfide (WS) to surpass this limit and realize highly reconfigurable, nonvolatile photoresponsivity with a retinomorphic photovoltaic device.

Although photodynamic immunotherapy represents a promising therapeutic approach against malignant tumors, its efficacy is often hampered by the hypoxia and immunosuppressive conditions within the tumor microenvironment (TME) following photodynamic therapy (PDT). In this study, we report the design guidelines towards efficient Type-I semiconducting polymer photosensitizer and modify the best-performing polymer into a hypoxia-tolerant polymeric photosensitizer prodrug (HTPS) for cancer photo-immunotherapy. HTPS not only performs Type-I PDT process to partially overcome the limitation of hypoxic tumors in PDT by recycling oxygen but also specifically releases a Signal Transducer and Activator of Transcription-3 (STAT3) inhibitor (Niclosamide) in response to a cancer biomarker in the TME.

Acylation stands as a fundamental process in both biological pathways and synthetic chemical reactions, with acylated saccharides and their derivatives holding diverse applications ranging from bioactive agents to synthetic building blocks. A longstanding objective in organic synthesis has been the site-selective acylation of saccharides without extensive pre-protection of alcohol units. In this study, we demonstrate that by simply altering the chirality of N-heterocyclic carbene (NHC) organic catalysts, the site-selectivity of saccharide acylation reactions can be effectively modulated.

Enzyme-catalyzed protein modifications have become invaluable in diverse applications, outperforming chemical methods in terms of precision, conjugation efficiency, and biological compatibility. Despite significant advances in ligases, such as sortase A and OaAEP1, their use in heterogeneous biological environments remains constrained by limited target sequence specificity. Methods: To test these hypotheses in Ophiorrhiza japonica, we compared pollinator visitation frequencies and plant traits between pink and white morphs in Shibing, a dolomite Karst region recognized as a World Natural Heritage Site.

Low-dimensional lead-free metal halide perovskites are highly attractive for cutting-edge optoelectronic applications. Herein, we report a class of scandium-based double perovskite crystals comprising antimony dopants that can generate multiexcitonic emissions in the ultraviolet, blue, and yellow spectral regions. Owing to the zero-dimensional nature of the crystal lattice that minimizes energy crosstalk, different excitonic states in the crystals can be selectively excited by ultraviolet light, X-ray irradiation, and mechanical action, enabling dynamic control of steady/transient-state spectral features by modulating the excitation modes.

Objective: To pilot a digital health technologies ecosystem known as project SingaporeWALK (earables and pps for ommunity iving and nowledge) that build capacity in older adults, senior center managers, health coaches, and caregivers in using health technologies (eg, wearables, apps, exergames) collaboratively in a gamified way for active aging.
Materials And Methods: The SingaporeWALK ecosystem was set up through 3 initiatives: (1) co-developing technologies with stakeholders; (2) raising digital literacy and capacity building; and (3) cultivating community and intergenerational bonding for active aging through gamified technology use.
Results: Significant improvements in older adults' self-reported physical and mental health post-intervention were observed.
