Pluripotent stem cells possess a unique nuclear architecture characterized by a larger nucleus and more open chromatin, which underpins their ability to self-renew and differentiate. Here, we show that the nucleolus-specific RNA helicase DDX18 is essential for maintaining the pluripotency of human embryonic stem cells. Using techniques such as Hi-C, DNA/RNA-FISH, and biomolecular condensate analysis, we demonstrate that DDX18 regulates nucleolus phase separation and nuclear organization by interacting with NPM1 in the granular nucleolar component, driven by specific nucleolar RNAs.

Owning to the versatile nature in participation of Diels-Alder (D-A) reactions, the development of efficient approaches to generate active ortho-quinodimethanes (o-QDMs) has gained much attention. However, a catalytic method involving coupling of two readily accessible components to construct o-QDMs is lacking. Herein, we describe a palladium carbene migratory insertion enabled dearomative C(sp)-H activation to form active o-QDM species through the cross-coupling of N-tosylhydrazones with aryl halides.

By targeting the essential viral RNA-dependent RNA polymerase (RdRP), nucleoside analogs (NAs) have exhibited great potential in antiviral therapy for RNA virus-related diseases. However, most ribose-modified NAs do not present broad-spectrum features, likely due to differences in ribose-RdRP interactions across virus families. Here, we show that HNC-1664, an adenosine analog with modifications both in ribose and base, has broad-spectrum antiviral activity against positive-strand coronaviruses and negative-strand arenaviruses.

A novel straightforward synthetic route to monocyclic 1,3,2,4-diazadiborinines has been developed by the sequential reaction of the NHC-coordinated iminoborane with bases and haloboranes (or borate). The first examples of monocyclic 1,3,2,4-diazadiborinines featuring different functional groups on the two B atoms have been synthesized and structurally characterized. Further derivatization of 4-bromophenyl-substituted 1,3,2,4-diazadiborinine has also been achieved, giving the biphenyl-substituted 1,3,2,4-diazadiborinine.

Developing miniaturized low-frequency acoustic sensors with high sensitivity is crucial for diverse applications, including geological monitoring and aerospace exploration. However, the performance of low-frequency acoustic sensor is constrained by the limited mechanical robustness of traditional sensing films at nanoscale thickness. Here, a functionalized graphene oxide (GO)-based Fabry-Perot (FP) low-frequency sensor is proposed, with characteristics of compact size, resistance to electromagnetic interference high-sensitivity low minimum detectable pressure (MDP), and a high signal-to-noise ratio (SNR).

Mammalian SWI/SNF (mSWI/SNF) ATP-dependent chromatin remodeling complexes play critical roles in regulating gene expression and DNA accessibility, and more than 20 % of cancers have mutations in genes encoding chromatin remodeling complexes. The mSWI/SNF family comprises three distinct classes: canonical BAF (cBAF), PBAF, and non-canonical BAF (ncBAF). While the structures of cBAF and PBAF have been resolved by using cryo-electron microscopy (cryo-EM), the modular organization and assembly mechanism of ncBAF remain poorly understood.
