The structures of - were elucidated by detailed analysis of their spectroscopic data, and the known compounds - were identified by comparison of their spectroscopic data with those reported. Since argentatin C () was found to exhibit antinociceptive activity by decreasing the excitability of rat and macaque dorsal root ganglia (DRG) neurons, and its new analogues - were evaluated for their ability to decrease the excitability of rat DRG neurons.

In this perspective, we outline a new opportunity for exploiting nanoparticle delivery of antagonists to target G-protein coupled receptors localized in intracellular compartments. We discuss the specific example of antagonizing endosomal receptors involved in pain to develop long-lasting analgesics but also outline the broader application potential of this delivery approach. We discuss the materials used to target endosomal receptors and indicate the design requirements for future successful applications.

Phototherapy is an emerging non-pharmacological treatment for depression, circadian rhythm disruptions, and neurodegeneration, as well as pain conditions including migraine and fibromyalgia. However, the mechanism of phototherapy-induced antinociception is not well understood. Here, using fiber photometry recordings of population-level neural activity combined with chemogenetics, we found that phototherapy elicits antinociception via regulation of the ventral lateral geniculate body (vLGN) located in the visual system.

The hypothesis that sustained G protein-coupled receptor (GPCR) signaling from endosomes mediates pain is based on studies with endocytosis inhibitors and lipid-conjugated or nanoparticle-encapsulated antagonists targeted to endosomes. GPCR antagonists that reverse sustained endosomal signaling and nociception are needed. However, the criteria for rational design of such compounds are ill-defined.

Objectives: The authors aim to evaluate a scale assessing oral health knowledge, the KROHL (Knowledge Related to Oral Health Literacy) including the inter-rater reliability for scoring open-ended questions, internal consistency of the hypothesized scales, the discriminant validity of the resulting scale and its relationship to existing measures of oral health literacy.
Methods: The questionnaire was administered via face-to-face interviews to 144 volunteers recruited in the waiting areas of clinics throughout the NYU College of Dentistry.The KROHL questionnaire evaluates oral health knowledge by asking open-ended questions about the appearance, cause, treatment and prevention of caries, gum disease, oral cancer, tooth loss and malocclusion.

Methods: Mandibular primary molar crowns fabricated by experimental (EB) or commercially available CAD/CAM RCs (HC) were prepared and cemented to a resinous abutment tooth using an adhesive resin cement (Cem) or a conventional glass-ionomer cement (CX). These specimens were subjected to a single compressive test (n = 5/each) and the step-stress accelerated life testing (SSALT) (n = 12/each).

Objective: The objective of this study was to manufacture and to evaluate the physico-mechanical properties of the Lithium disilicate (LiOSi) ceramic structures fabricated using additive manufacturing (3D printing).
Methods: LiOSi samples were divided into (n = 30/group): SM (subtractively manufactured) and AR (additive/robocasting). For the AR group, LiOSi powder was combined with ammonium polyacrylate, hydroxypropyl methylcellulose, and polyelectrolyte to create a colloidal gel, which was then used for printing.

The vasopressin type 2 receptor (VR) is an essential GPCR in renal regulation of water homeostasis. Upon stimulation, the VR activates Gα and Gα, which is followed by robust recruitment of β-arrestins and receptor internalization into endosomes. Unlike canonical GPCR signaling, the β-arrestin association with the VR does not terminate Gα activation, and thus, Gα-mediated signaling is sustained while the receptor is internalized.
