Studies have linked inhalation anesthesia and surgery to increased cognitive impairment, particularly in the elderly. Our previous research showed that isoflurane, but not desflurane, affected cognitive function in mice by modulating cyclophilin D (CypD), a key regulator of the mitochondrial permeability transition pore (mPTP) and mitochondrial function. Both anesthetics metabolize into trifluoroacetic acid (TFA), which is associated with cognitive deficits.

Leukemia-driving mutations are thought to arise in hematopoietic stem cells (HSC), yet the natural history of their spread is poorly understood. We genetically induced mutations within endogenous murine HSC and traced them in unmanipulated animals. In contrast to mutations associated with clonal hematopoiesis (such as Tet2 deletion), the leukemogenic KrasG12D mutation dramatically accelerated HSC contribution to all hematopoietic lineages.

The effects of abrasion on the heating performance of carbon nanotube (CNT)/epoxy composites were investigated in terms of Joule's heat, convective heat, and radiative heat under moderate-to-severe and localized abrasive conditions. While the overall heating behavior was characterized by the heating rate and the curvature of the transient response, a numerical solution of the heat equation was used to quantify convective and radiative heat transfers, incorporating the specific heat of each component, the convective heat transfer coefficient, and the Biot number. Black women experience the highest mortality and morbidity resulting from intimate partner violence (IPV) victimization, yet there remains a dearth of culturally responsive interventions designed to meet their needs within the coordinated community response system. We employed the Theory of Help-Seeking Behavior to explicate the barriers that Black women experience when securing assistance from providers within the IPV service provision system, inclusive of the criminal legal, child protective service, shelter, healthcare, and mental healthcare systems. In-depth individual interviews were conducted with 30 people who self-identified as Black women who were help-seeking within the IPV service provision system at the time of data collection.

Archival fixed tissues hold key insights into the evolutionary history of RNA viruses and the associated host immune response, yet access to the RNA sequence data is limited by a lack of robust methods for RNA extraction and sequence retrieval from these tissue types. Here we compared three commercial RNA extraction techniques (bead, column, and phase-based) on five fixed human brain tissues done in triplicate, that have been stored for up to 43 years. We found that for this sample set, bead-based extractions captured longer molecules and yielded a greater proportion of unique reads when aligned to the human genome, than did column and phase-based extraction methods.

The diagnosis, risk factors, treatment algorithms, and long-term sequelae of superficial and deep, implant-related infections in transdermal, bone-anchored osseointegration are not well-defined. In contrast to the robust experience diagnosing and managing periprosthetic joint infections in total joint arthroplasty, osseointegration surgery has only recently been adopted at a small number of osseointegration centers in the United States, contributing to the lack of long-term outcomes. Through the pooled experience from these osseointegration centers, we present a consensus statement on the perioperative management, incidence, treatment, and diagnostic workup for infectious complications following transdermal, bone-anchored osseointegration.
