TAD-SIE: sample size estimation for clinical randomized controlled trials using a Trend-Adaptive Design with a Synthetic-Intervention-Based Estimator.

Background: Phase-3 clinical trials provide the highest level of evidence on drug safety and effectiveness needed for market approval by implementing large randomized controlled trials (RCTs). However, 30-40% of these trials fail mainly because such studies have inadequate sample sizes, stemming from the inability to obtain accurate initial estimates of average treatment effect parameters.

Methods: To remove this obstacle from the drug development cycle, we present a new algorithm called Trend-Adaptive Design with a Synthetic-Intervention-Based Estimator (TAD-SIE) that powers a parallel-group trial, a standard RCT design, by leveraging a state-of-the-art hypothesis testing strategy and a novel trend-adaptive design (TAD).

Introduction: Health disparities experienced by lesbian, gay, bisexual, and transgender (LGBT) individuals have been partially attributed to healthcare professionals' lack of cultural competence in addressing their specific needs. Early diagnosis is critical for providing a timely window for effective therapy in pulmonary fibrosis (PF); however, achieving this remains a significant challenge. The distinct honeycombing patterns observed in computed tomography (CT) for the primary diagnosis of PF are typically only visible in patients with moderate to severe disease, often leading to missed opportunities for early intervention. Warming and cooling catalyse widespread temporal turnover in biodiversity.

Turnover in species composition through time is a dominant form of biodiversity change, which has profound effects on the functioning of ecological communities. Turnover rates differ markedly among communities, but the drivers of this variation across taxa and realms remain unknown. Here we analyse 42,225 time series of species composition from marine, terrestrial and freshwater assemblages, and show that temporal rates of turnover were consistently faster in locations that experienced faster temperature change, including both warming and cooling.

Background: Elemental analysis of teeth allows for exposure assessment during critical windows of development and is increasingly used to link early life exposures and health. The measurement of inorganic elements in teeth is challenging; laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) is the most widely used technique.

Objective: Both synchrotron x-ray fluorescence (SXRF) and LA-ICP-MS have the capability to measure elemental distributions in teeth with each having distinct advantages and disadvantages.

Purpose: The purpose of this study is to analyze the trends and characteristics of ocular trauma research published from 2000 to 2022 to delineate the trajectory of the field's research, provide information about the network of key contributors, and help determine future research strategies and direction.

Methods: Web of Science was queried for published works using a series of keywords relating to ocular trauma: "globe rupture", "ruptured globe", "globe injury", "ocular trauma", "intraocular foreign body", "eye trauma", "eye injury", and "traumatic endophthalmitis". All article information was compiled using the VOSviewer software.

The dynamic landscape of mRNA technology highlights the need for innovative quality control (QC) strategies. In this study, we described an efficient one-step digestion approach for concurrent generation of 5'- and 3'-end fragments, enabling simultaneous mRNA capping and poly(A) tail analysis. Tailored 10-23-type DNAzymes, designed from 5'- and 3'-Untranslated Regions (UTRs), selectively cleaved mRNA to release both the 5'-Capped or uncapped short fragments and 3'-Poly(A) tail cleavage products.

Effect of lemborexant on sleep architecture in participants with insomnia disorder and mild obstructive sleep apnea.

COMISA may negatively impact sleep duration and reduce rapid eye movement (REM) sleep, thereby impairing cognition. These post-hoc analyses evaluated the effect of lemborexant (LEM), a dual-orexin-receptor antagonist approved for adults with insomnia, on sleep architecture in participants with COMISA.

Anomalous superconductivity in twisted MoTe nanojunctions.

Introducing superconductivity in topological materials can lead to innovative electronic phases and device functionalities. Here, we present a unique strategy for quantum engineering of superconducting junctions in moiré materials through direct, on-chip, and fully encapsulated 2D crystal growth. We achieve robust and designable superconductivity in Pd-metalized twisted bilayer molybdenum ditelluride (MoTe) and observe anomalous superconducting effects in high-quality junctions across ~20 moiré cells.

REACH assessment of humans exposed to chemicals indirectly via the environment: screening modeling in EUSES versus state of the science.

The assessment of humans indirectly exposed to chemicals via the environment (HvE) is an assessment element of the Registration, Evaluation, Authorisation, and Restriction of Chemicals (REACH) regulation. The European Union System for the Evaluation of Substances (EUSES) is the default screening tool, aimed at prioritizing chemicals for further refinement/higher tier assessment. This review summarizes the approach used in EUSES, evaluates the state of the science in human exposure modeling via the environment, and identifies areas for further research to strengthen the confidence and applicability of EUSES for assessing HvE.

Integrating ex situ biomimetic extraction analyses into contaminated sediment assessment and management decisions.

This study evaluated a novel ex situ passive sampling biomimetic extraction (BE) method to estimate toxic potency in sediments. Gas chromatography with flame ionization detection (GC-FID) analysis of polydimethylsiloxane fibers equilibrated with field sediments was used to quantify bioavailable polyaromatic hydrocarbons (PAHs) and other unresolved, site-specific contaminant mixtures. This method is biomimetic because contaminants partition to the fiber based on hydrophobicity and abundance, and GC-FID quantification accounts for all constituents absorbed to the fiber that may contribute to toxicity.

Motor evoked potentials (MEPs) are an important measure in transcranial magnetic stimulation (TMS) when assessing neuronal excitability in clinical diagnostics related to motor function, as well as in neuroscience research. However, manual feature extraction from large datasets can be time-consuming and prone to human error, and valuable features, such as MEP polyphasia and duration, are often neglected. Introduction: High-grade cervical intraepithelial neoplasia (CIN) is a premalignant lesion of the cervix caused by persistent human papillomavirus (HPV) infection, which can lead to cervical cancer. Despite ongoing primary prevention efforts, considerable burden of illness remains. This study assessed the epidemiological, economic, and humanistic burden associated with high-grade CIN among adult women in Europe.

Effects of tributyltin on placental and reproductive abnormalities in offspring.

Tributyltin (TBT) is an organotin compound and a common persistent environmental pollutant with endocrine-disrupting chemical (EDC) actions. It can accumulate in the environment at various concentrations throughout the food chain in the ecosystem, posing a risk to human health, especially during critical periods such as gestation and fetal and offspring development. In this review, we report the results of studies describing the consequences of TBT exposure on placental and reproductive parameters in offspring of both sexes.
