For instance, proper protein glycosylation, crucial for mAb stability, safety, and efficacy, heavily relies on cell culture conditions.

Microfluidic Cultures of Basal Forebrain Cholinergic Neurons for Assessing Retrograde Cell Death by Live Imaging.

Neurons are highly polarized cells, with axons that may innervate distant target regions. In the brain, basal forebrain cholinergic neurons (BFCNs) possess extensive axons that project to several target regions such as the cortex, hippocampus, and amygdala, and may be exposed to a specific microenvironment in their axon targets that may have retrograde effects on neuronal health. Interestingly, BFCNs express the pan-neurotrophin receptor p75NTR throughout life while also concomitantly co-expressing all Trk receptors, making them capable of responding to both mature and precursor neurotrophins to promote survival or apoptosis, respectively.

Optic capture of a three-piece lens in the absence of posterior capsule support is an effective strategy for stabilizing and sequestering the optic to reduce the risk of dislocation and UGH syndrome compared to sulcus placement. We present a novel technique that facilitates optic capture in the presence of a contracted, fibrotic rhexis opening, while minimizing stress on the zonules by using iris retractors to assist in stabilization and expansion of the rhexis, followed by direct injection of the optic of the lens behind the anterior capsule opening into an optic captured configuration.

Connectivity changes following transcranial alternating current stimulation at 5-Hz: an EEG study.

Transcranial alternating current stimulation (tACS) at 5-Hz to the right hemisphere can alleviate anxiety symptoms. We aimed to explore the connectivity changes following the treatment. We collected electroencephalography (EEG) data from 24 participants with anxiety disorders before and after the tACS treatment during a single session.

Tracing the change of the volatile compounds of soy sauce at different fermentation times by PTR-TOF-MS, -nose and GC-MS.

Proton-transfer reaction time-of-flight mass spectrometry (PTR-TOF-MS), combined with electronic nose (-nose), was first used to track the change of volatile organic compounds (VOCs) in soy sauce in this study. The results showed that 163 VOCs with different mass numbers were identified. Based on the differences in VOCs, the entire fermentation cycle was divided into four stages (0D and 15D; 30D-75D; 90D; 105D-120D).

Multicenter analysis of the incidence of in-stent stenosis following the deployment of flow-redirection endoluminal device in the treatment of intracranial aneurysms.

Introduction: The Flow Re-direction Endoluminal Device (FRED) is a novel flow diverter with a unique double stent design, with an inner stent composed of 48 nitinol wires, and an outer stent with 16 nitinol wires. It is designed for endovascular cerebral aneurysm treatment, although, limited data exist regarding in-stent stenosis (ISS) rates associated with FRED devices.

Methods: A registry encompassing two North American comprehensive stroke centers was the base of this study.

Causal inference with cross-temporal design.

When many participants in a randomized trial do not comply with their assigned intervention, the randomized encouragement design is a possible solution. In this design, the causal effects of the intervention can be estimated among participants who would have experienced the intervention if encouraged. For many policy interventions, encouragements cannot be randomized and investigators need to rely on observational data.

Safety, efficacy, and mechanism of action of the temporo-masseteric nerve block.

The objective of the study was to assess the utility and safety of Temporo-masseteric Nerve Block (TMNB), and to explore the mechanism for its apparent sustained pain relief. This manuscript describes, (1) a retrospective study evaluating pain reduction in patients who received the TMNB injection for the management of masticatory myogeneous pain (myalgia, per Diagnostic Criteria for Temporomandibular Disorders (DC/TMD criteria)), and (2) a motor nerve conduction study (NCS) of the temporalis and masseter, performed in the absence of signs or symptoms of TMD, before and after the TMNB injection. Orofacial migraine (OFM) and neurovascular orofacial pain (NVOP) are both recognized as migraine-related entities affecting the facial and orofacial regions, according to the International Classification of Orofacial Pain (ICOP). However, the distinction between these two conditions and the question of whether NVOP should be considered a separate entity remain subjects of ongoing debate. The aim of this study is to compare the diagnostic characteristics of OFM and NVOP to reassess whether they should continue to be classified as two distinct diagnoses.

Major individual and regional variations in unit entrainment by oscillations of different frequencies.

In vitro studies have shown that a neuron's electroresponsive properties can predispose it to oscillate at specific frequencies. In contrast, network activity in vivo can entrain neurons to rhythms that their biophysical properties do not predispose them to favor. However, there is limited information on the comparative frequency profile of unit entrainment across brain regions.

Autonomic features of craniofacial neuralgias: a systematic review with meta-analysis.

The aim of the study is to describe the severity, temporal characteristics, and types of autonomic features as they relate to the characteristics of pain of the neuralgias. Also, to describe, based on literature, how these autonomic features can affect the treatment outcomes of patients with craniofacial neuralgias. We carried out a literature search using five databases, PubMed, Embase, OVID, Scopus and Web of Science.

Background: Soluble species of multimeric amyloid-beta including globular amyloid-beta oligomers (AβOs) and linear amyloid-beta protofibrils are toxic to neurons. Sabirnetug (ACU193) is a humanized monoclonal antibody, raised against globular species of soluble AβO, that has over 650-fold greater binding affinity for AβOs over monomers and appears to have relatively little binding to amyloid plaque.

Objectives: To assess safety, pharmacokinetics, and exploratory measures including target engagement, biomarker effects, and clinical efficacy of sabirnetug in participants with early symptomatic Alzheimer's disease (AD; defined as mild cognitive impairment and mild dementia due to AD).

Key considerations for combination therapy in Alzheimer's clinical trials: Perspectives from an expert advisory board convened by the Alzheimer's drug discovery foundation.

There is growing consensus in the Alzheimer's community that combination therapy will be needed to maximize therapeutic benefits through the course of the disease. However, combination therapy raises complex questions and decisions for study sponsors, from preclinical research through clinical trial design to regulatory, statistical, and operational considerations. In January 2024, the Alzheimer's Drug Discovery Foundation convened an expert advisory board to discuss the key considerations in each of these areas.

Compensation Study of Vascular Surgeons in the United States.

Objective: The Society for Vascular Surgery (SVS) partnered with Phairify, Inc, an organization with experience in physician compensation data compilation for several other medical specialties, to survey its membership and assess factors influencing vascular surgeon compensation.

Methods: The SVS Compensation Study Task Force developed a vascular surgery-specific survey between January 2023 and May 2023 including experience level, academic rank, bonuses, incentives, gender, race, ethnicity, geography, on-call pay, and other factors influencing overall reimbursement. After a soft launch on May 1, 2023, with an initial phase of SVS leadership engagement in completion, the survey was formally introduced to the SVS membership on June 14, 2023.
